Chronic Methamphetamine Effects on Brain Structure and Function in Rats

Methamphetamine (MA) addiction is a growing epidemic worldwide. Chronic MA use has been shown to lead to neurotoxicity in rodents and humans. Magnetic resonance imaging (MRI) studies in MA users have shown enlarged striatal volumes and positron emission tomography (PET) studies have shown decreased brain glucose metabolism (BGluM) in the striatum of detoxified MA users. The present study examines structural changes of the brain, observes microglial activation, and assesses changes in brain function, in response to chronic MA treatment. Rats were randomly split into three distinct treatment groups and treated daily for four months, via i.p. injection, with saline (controls), or low dose (LD) MA (4 mg/kg), or high dose (HD) MA (8 mg/kg). Sixteen weeks into the treatment period, rats were injected with a glucose analog, [F-18] fluorodeoxyglucose (FDG), and their brains were scanned with micro-PET to assess regional BGluM. At the end of MA treatment, magnetic resonance imaging at 21T was performed on perfused rats to determine regional brain volume and in vitro [H-3]PK 11195 autoradiography was performed on fresh-frozen brain tissue to measure microglia activation. When compared with controls, chronic HD MA-treated rats had enlarged striatal volumes and increases in [H-3]PK 11195 binding in striatum, the nucleus accumbens, frontal cortical areas, the rhinal cortices, and the cerebellar nuclei. FDG microPET imaging showed that LD MA-treated rats had higher BGluM in insular and somatosensory cortices, face sensory nucleus of the thalamus, and brainstem reticular formation, while HD MA-treated rats had higher BGluM in primary and higher order somatosensory and the retrosplenial cortices, compared with controls. HD and LD MA-treated rats had lower BGluM in the tail of the striatum, rhinal cortex, and subiculum and HD MA also had lower BGluM in hippocampus than controls. These results corroborate clinical findings and help further examine the mechanisms behind MA-induced neurotoxicity.

Engineering extracellular vesicles by three-dimensional dynamic culture of human mesenchymal stem cells

Summary: This study presents a promising strategy for scalable production of high-quality extracellular vesicles (EVs) from human mesenchymal stem cells (hMSCs) using 3D aggregates grown under wave motion. The 3D hMSC aggregates promote activation of different pathways involved in EV secretion. Comparison with 2D hMSC-EVs reveals that the quantity of 3D hMSC-EVs is significantly enhanced, with smaller sizes, higher expression of miR-21 and miR-22, and an altered protein cargo. Functionally, 3D hMSC-EVs rejuvenate senescent stem cells and exhibit enhanced immunomodulatory potentials.

Abstinence-Dependent Effects of Long-Access Cocaine Self-Administration on Nucleus Accumbens Astrocytes Are Observed in Male, But Not Female, Rats

NeuroConstruct: 3D Reconstruction and Visualization of Neurites in Optical Microscopy Brain Images

Human Forebrain Organoid-Derived Extracellular Vesicle Labeling with Iron Oxides for In Vitro Magnetic Resonance Imaging

Superparamagnetic iron oxide (SPIO) nanoparticles have been shown to improve the targeting ability of EVs. In this study, ultrasound sonication was used to load ultrasmall SPIO (USPIO) nanoparticles onto EVs derived from induced forebrain neural progenitor cortical organoids (iNPCo). The labeled EVs exhibited higher MRI contrast in agarose gel phantoms.

NeuRegenerate: A Framework for Visualizing Neurodegeneration

Summary: Recent advances in high-resolution microscopy have enabled scientists to study brain connectivity and predict changes in neural fiber morphology over time using a novel framework called NeuRegenerate. This framework incorporates a deep-learning network called neuReGANerator, which uses a cycle-consistent generative adversarial network (GAN) to translate features of neuronal structures across age-timepoints. The framework also includes visualization tools, such as neuroCompare and neuroMorph, to visualize the differences and transformations in neural projections. The framework is specifically designed for wide-field microscopy volumes and has been demonstrated in the study of cholinergic system changes in mouse brains.

Probing self-assembled micellar topologies via micro-scale diffusive dynamics of surfactants
