Fabrication of Acrylonitrile-Butadiene-Styrene Nanostructures with Anodic Alumina Oxide Templates, Characterization and Biofilm Development Test for Staphylococcus epidermidis

Medical devices can be contaminated by microbial biofilm which causes nosocomial infections. One of the strategies for the prevention of such microbial adhesion is to modify the biomaterials by creating micro or nanofeatures on their surface. This study aimed (1) to nanostructure acrylonitrile-butadiene-styrene (ABS), a polymer composing connectors in perfusion devices, using Anodic Alumina Oxide templates, and to control the reproducibility of this process; (2) to characterize the physico-chemical properties of the nanostructured surfaces such as wettability using captive-bubble contact angle measurement technique; (3) to test the impact of nanostructures on Staphylococcus epidermidis biofilm development. Fabrication of Anodic Alumina Oxide molds was realized by double anodization in oxalic acid. This process was reproducible. The obtained molds present hexagonally arranged 50 nm diameter pores, with a 100 nm interpore distance and a length of 100 nm. Acrylonitrile-butadiene-styrene nanostructures were successfully prepared using a polymer solution and two melt wetting methods. For all methods, the nanopicots were obtained but inside each sample their length was different. One method was selected essentially for industrial purposes and for better reproducibility results. The flat ABS surface presents a slightly hydrophilic character, which remains roughly unchanged after nanostructuration, the increasing apparent wettability observed in that case being explained by roughness effects. Also, the nanostructuration of the polymer surface does not induce any significant effect on Staphylococcus epidermidis adhesion.

Understanding and Characterizing the Drug Sorption to PVC and PE Materials

Summary: Characterizing the sorption of drugs onto polyvinylchloride (PVC) and polyethylene (PE) materials in terms of thermodynamic adsorption properties and atomistic details is crucial for developing alternative materials that can reduce drug sorption phenomena and plasticizer release. Experiments and sophisticated calculations were used to study the sorption of paracetamol and diazepam on PE and PVC surfaces, with simulated Gibbs free energies of adsorption matching experimental interpretations. Furthermore, a molecular-scale characterization of the polymer-drug-water interface was conducted, exploring local properties such as density, orientation, and diffusion.

Association between Urinary Metabolites and the Exposure of Intensive Care Newborns to Plasticizers of Medical Devices Used for Their Care Management

Infusion sets in neonatology: What practices in France?

In vitro and in silico approach to study the hormonal activities of the alternative plasticizer tri-(2-ethylhexyl) trimellitate TEHTM and its metabolites

Summary: This study investigated the endocrine-disrupting effects of metabolites of the plasticizer TEHTM, commonly used in medical devices. The results showed that two of the metabolites (4-MEHTM and 2/1-MEHTM) had effects on hormone activity in vitro. However, the concentrations of these metabolites found in human urine were unlikely to have active effects on estrogen, androgen, thyroid receptors, and steroidogenesis-mediated effects.

Cyclodextrins Allow the Combination of Incompatible Vancomycin and Ceftazidime into an Ophthalmic Formulation for the Treatment of Bacterial Keratitis

Summary: Ceftazidime and vancomycin are used to treat bacterial keratitis, but their physical incompatibility prevents them from being mixed in a single container for ophthalmic administration. Researchers investigated the use of cyclodextrins to prevent this incompatibility and determined optimal conditions for a formulation with low turbidity and particle count. The formulation was found to inhibit incompatibility at pH levels above 7.3 while maintaining antibacterial activity.

Critical Drug Loss Induced by Silicone and Polyurethane Implantable Catheters in a Simulated Infusion Setup with Three Model Drugs

Summary: The study experimentally investigated the drug loss caused by silicone and polyurethane implantable catheters, finding that polyurethane catheters caused the highest losses of diazepam and insulin. Drug concentrations were monitored by liquid chromatography, with no loss of paracetamol observed in the complete setups. Catheters were shown to be the cause of some extremely high drug losses that could not be countered by optimizing the extension set in the setup.

Physicochemical Stability of a Novel Tacrolimus Ophthalmic Formulation for the Treatment of Ophthalmic Inflammatory Diseases

Summary: This study evaluated the physicochemical stability of a preservative-free tacrolimus formulation at different storage temperatures. The results showed that the 0.2 mg/mL and 1 mg/mL tacrolimus ophthalmic solutions remained stable when stored at 5 degrees Celsius, but signs of chemical instability were observed when stored at 25 degrees Celsius and 35 degrees Celsius.

Ex Vivo Model to Assess the Exposure of Patients to Plasticizers from Medical Devices during Pre-CAR-T Cells' Apheresis

Summary: This study aimed to evaluate the migration of plasticizers during a pre-CAR-T cell leukapheresis procedure and the exposure of patients and their lymphocytes to them. The results showed that DEHP was present in the leukapheresis kit, with migration levels ranging from 25% to 59%. A total of 98.90±11.42 mg of DEHP was released into the simulant. This exposure could potentially impact the health of patients and the efficiency of CAR-T cells.

On-call duties in hospital pharmacies: National survey and elaboration of a training program for pharmacy residents

Summary: Our study aimed to develop and optimize a structured and harmonized training program for pharmacy residents participating in hospital on-call duties, in response to disparities and lack of structure in training observed nationwide. The program consists of theoretical, practical, and continuous training, with the introduction of tools such as in-house on-call duty notebook, simulation workshops, and interactive quizzes to enhance training quality and effectiveness. Collaboration with pharmacists from different sectors facilitated the gradual implementation of the training in our hospital pharmacy.
