Mitogenomes from The 1000 Genome Project Reveal New Near Eastern Features in Present-Day Tuscans

Background Genetic analyses have recently been carried out on present-day Tuscans (Central Italy) in order to investigate their presumable recent Near East ancestry in connection with the long-standing debate on the origins of the Etruscan civilization. We retrieved mitogenomes and genome-wide SNP data from 110 Tuscans analyzed within the context of The 1000 Genome Project. For phylogeographic and evolutionary analysis we made use of a large worldwide database of entire mitogenomes (>26,000) and partial control region sequences (>180,000). Results Different analyses reveal the presence of typical Near East haplotypes in Tuscans representing isolated members of various mtDNA phylogenetic branches. As a whole, the Near East component in Tuscan mitogenomes can be estimated at about 8%; a proportion that is comparable to previous estimates but significantly lower than admixture estimates obtained from autosomal SNP data (21%). Phylogeographic and evolutionary inter-population comparisons indicate that the main signal of Near Eastern Tuscan mitogenomes comes from Iran. Conclusions Mitogenomes of recent Near East origin in present-day Tuscans do not show local or regional variation. This points to a demographic scenario that is compatible with a recent arrival of Near Easterners to this region in Italy with no founder events or bottlenecks.

Summary: This study used a cardiac gene panel for sequencing and ExomeDepth software for copy number variation (CNV) detection. After a rigorous prioritization process, three real CNVs were confirmed in five individuals in the MYH11, FBN1, and PDMI7 genes. Based on Bayes factors, BF values above 60 were established as the threshold for considering real CNVs in the calling process carried out by ExomeDepth on our gene panel.

Sensogenomics of music and Alzheimer's disease: An interdisciplinary view from neuroscience, transcriptomics, and epigenomics

Summary: This study systematically reviewed the state-of-the-art of neuroscience and cognitive sciences research on music and Alzheimer's disease (AD) and found that music has beneficial effects on patients with neurodegenerative diseases, especially in memory and cognition. Transcriptomic and epigenomic data analysis also revealed connections between music-related genes and AD. These findings provide a new roadmap for genetic research and musical interventions in AD and other neurodegenerative conditions.

Summary: This study investigated febrile children with petechial rashes who presented to European emergency departments and found that children with a combination of fever and petechial rash were more likely to have sepsis, meningitis, bacterial infections, and require immediate life-saving interventions and intensive care unit admissions. Therefore, the combination of fever and petechial rash is still an important warning sign for childhood sepsis and meningitis.

The study found that children with prolonged fever have a higher risk of serious bacterial infection (SBI). However, warning signs of SBI were observed infrequently, highlighting the need for careful clinical assessment and diagnostic workup.

Summary: This study estimated the clinical and economic burden of severe influenza in Spain using statistical models. The results showed that there is a significant under-detected burden of influenza, especially in the elderly population. The average annual cost of influenza-related hospitalizations was 45.7 million euros, and the average annual mortality rate due to influenza was 27.7 per 100,000 people.

Summary: Universal immunization greatly reduces the incidence and mortality of vaccine-preventable diseases. However, routine immunization coverage varies significantly between countries, populations, and districts, and has even declined in some cases. Insufficient immunization coverage leads to susceptible individuals and outbreaks of vaccine-preventable diseases. The European Immunization Agenda 2030 aims to promote equity in immunization and assist stakeholders in addressing local challenges to improve overall health in the WHO European Region.

Summary: This study investigated the presence of anticardiac autoantibodies in MIS-C or COVID-19 vaccine-induced myocarditis and found no evidence of antibody binding to cardiac tissue in these conditions. This suggests that the cardiac pathology in MIS-C and vaccine-induced myocarditis is unlikely to be driven by direct anticardiac antibody-mediated mechanisms.

Chatting with ChatGPT to learn about safety of COVID-19 vaccines - A perspective

This study investigates the ability of the AI chatbot ChatGPT to generate opinions on vaccine hesitancy by examining its response to prevalent false information, contraindications, and myths about vaccine safety circulating on the internet. The findings suggest that while ChatGPT's default responses may be incomplete, they are generally satisfactory and have the potential to guide users towards scientifically aligned information, though it cannot replace experts or scientific evidence itself.

Impact of infection on proteome-wide glycosylation revealed by distinct signatures for bacterial and viral pathogens

Summary: Mechanisms of infection and pathogenesis have mainly focused on differential gene or protein expression, with less emphasis on posttranslational modifications. In this study, a novel glycoproteomics approach was applied to investigate systemic proteome-wide glycosylation in response to infection. Through characterization of site-specific protein glycosylation in plasma samples from controls and patients, a glycopeptide signature was identified that could significantly differentiate between bacterial and viral infections. Additionally, machine learning algorithm-based analysis demonstrated the ability to identify the causative pathogens based on distinct host blood plasma glycopeptide signatures. These findings highlight the potential of glycoproteomics as an innovative approach to enhance the interpretation of relevant biological changes during infection.
