Summary: This study aimed to analyze the differential gene expression in XP-A cells after UV irradiation. The results showed that the number of genes with significantly altered expression in XP-A cells at 12 hours was higher compared to normal cells, indicating a correlation between the number of altered genes and DNA damage. The study found that the XPA protein can transcriptionally inhibit replication-related genes and regulate DNA replication and re-replication after UV irradiation.

Summary: We synthesized a new tubulin polymerization inhibitor, RS6077, which inhibited the growth of multiple cancer cell lines without affecting non-transformed cells. It induced G2/M phase arrest in both transformed and non-transformed cells, but showed selective induction of cell death in transformed cells that fail to complete mitosis. RS6077 also inhibited the growth of a lymphoma xenograft model and demonstrated good metabolic stability.

Summary: R-loops are three-stranded structures formed by annealing of nascent transcripts and template DNA, playing important roles in physiological processes. Dysregulation of R-loop metabolism may pose a threat to genome stability. Depletion of RAD51 and inhibition by B02 did not prevent R-loop formation in human cells, while B02 treatment may lead to premature origin firing caused by RAD51-dependent accumulation of R-loops.

Summary: Research has shown that ExoT induces potent apoptotic cytotoxicity in cancer cell lines through two distinct mechanisms and significantly reduces tumor growth in melanoma. ExoT exhibits antiproliferative function in melanoma cells by causing cell cycle arrest in G1 interphase through dampening G1/S checkpoint proteins. Both domains of ExoT contribute to G1 cell cycle arrest in melanoma, with ADPRT likely involving the Crk adaptor protein.

Both qualitative and quantitative models of cell cycle control are reflected in budding yeast, suggesting the importance of cyclin specificity in cell cycle regulation.

Summary: Metastatic breast cancer (mBC) is responsible for >90% of breast cancer-related deaths. The effectiveness of microtubule-targeting agents (MTAs) in mBC treatment is limited by primary or acquired resistance. This study evaluated the potential of methyl N-(6-benzoyl-1H-benzimidazol-2-yl)carbamate (BCar) as a novel MTA for MTA-resistant mBC. The results showed that BCar selectively killed breast cancer cells and had a wider therapeutic window compared to clinical MTAs docetaxel and vincristine.
