Acidophilic Green Alga Pseudochlorella sp YKT1 Accumulates High Amount of Lipid Droplets under a Nitrogen-Depleted Condition at a Low-pH

Microalgal storage lipids are considered to be a promising source for next-generation biofuel feedstock. However, microalgal biodiesel is not yet economically feasible due to the high cost of production. One of the reasons for this is that the use of a low-cost open pond system is currently limited because of the unavoidable contamination with undesirable organisms. Extremophiles have an advantage in culturing in an open pond system because they grow in extreme environments toxic to other organisms. In this study, we isolated the acidophilic green alga Pseudochlorella sp. YKT1 from sulfuric acid mine drainage in Nagano Prefecture, Japan. The vegetative cells of YKT1 display the morphological characteristics of Trebouxiophyceae and molecular phylogenetic analyses indicated it to be most closely related to Pseudochlorella pringsheimii. The optimal pH and temperature for the growth of YKT1 are pH 3.0-5.0 and a temperature 20-25 degrees C, respectively. Further, YKT1 is able to grow at pH 2.0 and at 32 degrees C, which corresponds to the usual water temperature in the outdoors in summer in many countries. YKT1 accumulates a large amount of storage lipids (similar to 30% of dry weigh) under a nitrogen-depleted condition at low-pH (pH 3.0). These results show that acidophilic green algae will be useful for industrial applications by acidic open culture systems.

Cell size for commitment to cell division and number of successive cell divisions in cyanidialean red algae

Summary: The study reveals that there is a correlation between the number of successive cell divisions and the threshold cell size for cell division in different algae lineages, indicating that the threshold size for cell division plays a major role in determining the number of successive cell divisions. This suggests that evolutionary changes in the mechanism of cell size monitoring result in variations of multiple fission cell cycle in eukaryotic algae.

Three sex phenotypes in a haploid algal species give insights into the evolutionary transition to a self-compatible mating system

Summary: The study revealed a new haploid mating system in Pleodorina starrii with three sex phenotypes: male, female, and bisexual. The findings indicated that the strains belong to the same biological species, with intercrossings demonstrating normal zygote formation and high survivability of F1 strains.

Summary: The unicellular red alga Cyanidioschyzon merolae has a simple cellular and genomic architecture. A new nuclear targeting and transformant selection system using the antibiotic blasticidin S (BS) and the BS deaminase (BSD) selectable marker has been developed in C. merolae. This system enables simultaneous modification of two different chromosomal loci in the same strain, expanding the genetic modification capabilities of this alga.

Summary: Characteristics of transverse walls and modes of conjugation are crucial for species identification in the genus Spirogyra. This study examined two Spirogyra species with different types of transverse walls collected in Japan and found their distinct vegetative and reproductive traits. Molecular phylogenetic analyses also revealed their separate lineages in the tree of Spirogyra species. The two Japanese strains are described as new species, S. minuticrassispina sp. nov. and S. tertia sp. nov.

Life cycle and functional genomics of the unicellular red alga Galdieria for elucidating algal and plant evolution and industrial use

Summary: This study discovered that Galdieria exhibits a sexual life cycle, alternating between cell-walled diploid and cell wall-less haploid, while also having the ability for asexual reproduction. By studying the genome and transcriptome data of Galdieria, key transcription factors involved in cell transition and development were identified. Additionally, four actin genes were found to have different functions in motility and cytokinesis between the haploid and diploid stages, reflecting ancestral roles of actin. Furthermore, photosynthesis-deficient mutants were generated for potential industrial pigment production. Overall, these findings enhance our understanding of algae and plant evolution, as well as the industrial use of microalgae.

Expanded male sex-determining region conserved during the evolution of homothallism in the green alga Volvox

Summary: By comparing genome data from Thai and Japanese genotypes of the homothallic green alga Volvox africanus, it was found that expanded ancestral male and female sex-determining regions (SDRs) may have originated from an ancient heterothallic ancestor and were conserved during the evolution of homothallic genotypes. An expanded SDR-like region appears to be essential for homothallic sexual reproduction in V. africanus, regardless of its male or female origin. This study highlights the future possibility of elucidating the biological significance of these expanded genomic regions.
