The Expression of LEP, LEPR, IGF1 and IL10 in Obesity and the Relationship with microRNAs

Obesity is a multifactorial disease, with epigenetic alterations. Have been described modifications in the expression of some microRNAs, and some proteins related to obesity. The objective was to determine and correlate, in obese patients, the gene expression of LEP, LEPR, IGF1, IL10 and of miR-27a, miR-27b, miR-143 and miR-145. RNA was extracted from biopsies of subcutaneous fat, liver and visceral fat of 15 obese subjects submitted to bariatric surgery and of 15 non-obese subjects submitted to cholecystectomy for cDNA synthesis and for RT-PCR. The microRNAs were chosen using the TargetScan software. An increased expression of LEP and IGF1 was detected in the subcutaneous fat of the obese group compared to control, while the expression of IGF1 was higher in the control group than in the obese one. MiRNA-27a had a higher expression in the omentum of the obese patients and there was also a correlation in the expression of miRNA-145 and LEPR in the omentum of this group.

Summary: This study evaluated intestinal HLA-G expression and sHLA-G levels in CD/UC patients, finding increased HLA-G expression in infiltrating plasma cells/lymphocytes in CD/UC biopsies and higher levels of sHLA-G in lesion areas of surgical specimens from CD/UC patients compared to margins.Higher levels of sHLA-G were observed in UC/CD patients compared to controls, with no difference between the two diseases.

Summary: miRNAs were downregulated in both corpus cavernosum and blood in diabetic animals with alcohol consumption, showing potential as biomarkers for endothelial dysfunction. Serum miRNAs demonstrated high diagnostic accuracy in predicting endothelial dysfunction, suggesting their role as potential diagnostic biomarkers for endothelial injury.

Summary: Medulloblastoma in adults can be classified into WNT, SHH, Group 3, and Group 4 subgroups, with SHH tumors being diagnosed at an older age. The 5-year progression-free survival varies among different subtypes, but clinical variables do not show a significant association with subtype-specific PFS.

Summary: This study aimed to identify a tumor malignancy signature associated with the regulation of HIF-1 alpha by microRNAs miR-21 and miR-326 in glioblastoma stem cells. The results showed that miR-21 had a high level of expression in the irradiated neurosphere group, suggesting its role in radioresistant neurospheres. miR-326 was associated with tumor recurrence in both groups, indicating that its positive regulation is independent of ionizing radiation treatment.

Summary: Stroke is a major cause of death and disability worldwide. Physical exercise is considered an important approach for preventing non-communicable chronic diseases. miRNAs and other biomarkers respond to physical exercise and are associated with various epigenetic mechanisms. Studying the molecular mechanisms of physical exercise can improve the understanding of stimuli and optimal dosages, leading to better control of treatment and recovery processes for various diseases.
