Discovery of Specific Metastasis-Related N-Glycan Alterations in Epithelial Ovarian Cancer Based on Quantitative Glycomics

Generally, most of ovarian cancer cannot be detected until large scale and remote metastasis occurs, which is the major cause of high mortality in ovarian cancer. Therefore, it is urgent to discover metastasis-related biomarkers for the detection of ovarian cancer in its occult metastasis stage. Altered glycosylation is a universal feature of malignancy and certain types of glycan structures are well-known markers for tumor progressions. Thus, this study aimed to reveal specific changes of N-glycans in the secretome of the metastatic ovarian cancer. We employed a quantitative glycomics approach based on metabolic stable isotope labeling to compare the differential N-glycosylation of secretome between an ovarian cancer cell line SKOV3 and its high metastatic derivative SKOV3-ip. Intriguingly, among total 17 N-glycans identified, the N-glycans with bisecting GlcNAc were all significantly decreased in SKOV3-ip in comparison to SKOV3. This alteration in bisecting GlcNAc glycoforms as well as its corresponding association with ovarian cancer metastatic behavior was further validated at the glycotransferase level with multiple techniques including real-time PCR, western blotting, transwell assay, lectin blotting and immunohistochemistry analysis. This study illustrated metastasis-related N-glycan alterations in ovarian cancer secretome in vitro for the first time, which is a valuable source for biomarker discovery as well. Moreover, N-glycans with bisecting GlcNAc shed light on the detection of ovarian cancer in early peritoneal metastasis stage which may accordingly improve the prognosis of ovarian cancer patients.

Diagnostic Potential of Plasma IgG N-glycans in Discriminating Thyroid Cancer from Benign Thyroid Nodules and Healthy Controls

Summary: Novel biomarkers are urgently needed to distinguish between benign and malignant thyroid nodules and detect thyroid cancer in the early stage. The associations between serum IgG N-glycosylation and thyroid cancer risk have been revealed. Our results suggest that derived trait BN and the classification glyco-panel may serve as candidate biomarkers for further validation, showing accurate performance in discriminating thyroid cancer from benign thyroid nodules and healthy controls.

Transforming growth factor-β challenge alters the N-, O-, and glycosphingolipid glycomes in PaTu-S pancreatic adenocarcinoma cells

Summary: Transforming growth factor-beta (TGF-beta) plays a key role in pancreatic ductal adenocarcinoma (PDAC) progression, and glycosylation is associated with it. The study found that PDAC cells deficient in the signal transducer of TGF-beta signaling, SMAD4, exhibit glycosylation changes upon TGF-beta stimulation, including elevated N-glycome aberrations and alterations in O-glycosylation and glycosphingolipid glycosylation profiles.

Summary: This study demonstrated the potential diagnostic value of O-glycoforms in the hinge region of plasma IgA1 for IgA nephropathy. The levels of GalNAc and galactose were significantly different in IgA nephropathy patients compared to controls, and combining these values improved the diagnostic performance. A panel containing GalNAc, galactose, and circulating IgA showed excellent diagnostic potential.

Protein O-GlcNAcylation in cardiovascular diseases

Summary: O-GlcNAcylation is a protein post-translational modification controlled by O-GlcNAc transferase (OGT) and protein O-GlcNAcase (OGA), and is involved in the development of cardiovascular disease (CVD). Changes in O-GlcNAcylation levels can disrupt metabolic balance and affect cardiovascular function, but its role in CVD is still controversial. This review focuses on the interactions between protein O-GlcNAcylation and CVD and discusses potential molecular mechanisms for the treatment of cardiovascular events.

Comprehensive analysis of tissue proteomics in patients with papillary thyroid microcarcinoma uncovers the underlying mechanism of lymph node metastasis and its significant sex disparities

Summary: This study investigated the molecular mechanisms underlying lymph node metastasis (LNM) and its sex disparities in papillary thyroid microcarcinoma (PTMC) using a proteomics approach. The results showed that LNM in PTMC was associated with mitochondrial dysfunction and activation of oxidative phosphorylation and PI3K/AKT signaling pathways. The study also found distinct proteomics profiles between male and female PTMC patients with LNM, with elevated levels of extracellular matrix-associated proteins in male patients. Furthermore, certain proteins were identified to be downregulated and correlated with tumor metastasis, while others were upregulated and correlated with sex disparities and poor prognosis. These findings enhance our understanding of the functional networks and signaling pathways related to PTMC with LNM.

Serum protein N-glycome patterns reveal alterations associated with endometrial cancer and its phenotypes of differentiation

Summary: This study investigated the serum N-glycome patterns of endometrial cancer (EC) patients and found distinctive differences compared with healthy controls. Abnormal high-mannose and hybrid-type N-glycans, fucosylation, galactosylation, and linkage-specific sialylation were observed in EC patients. A glycan panel constructed with four discriminative traits accurately identified EC with high accuracy.

Summary: This study characterized the N-glycome of papillary thyroid microcarcinoma (PTMC), and established nomograms for the diagnosis of PTMC and the prediction of lymph node metastasis (LNM). The study found significant differences in serum N-glycome between PTMC and healthy controls, as well as associations between certain N-glycan traits and PTMC and LNM. Nomograms based on these traits showed good performance in the identification of PTMC and the prediction of LNM.
