Summary: Studies have shown that garlic extract (GE) can affect the expression of three genes (Ngn3, Pdx1, and MafA) in the pancreas and liver of diabetic rats. However, the effect of GE on the gene expression varied depending on the duration of treatment, with an initial increase followed by a decrease in expression. In addition, only Pdx1 expression increased significantly in the liver.

Summary: The study examines the relationship between DNA methylation and liver to pancreas transdifferentiation. The results show that the transdifferentiation process is associated with DNA demethylation and increased expression of specific genes.

Summary: This study used a label-free quantitative proteomic approach to explore the impact of a high-fat diet on liver tissue proteomic profile and identify novel transcription factors (TFs) related to nonalcoholic fatty liver disease (NAFLD). Results showed that metabolic-related proteins were significantly enriched and a subgroup of 11 TFs were found to interact with metabolic-related enzymes and kinases. Further analysis identified Rbbp4, Tcea1, and ILF2 as TFs that play important roles in lipid metabolism and NAFLD prevention.

Summary: This study demonstrates that coexpression of early temporal identity transcription factors Ikzf1 and Ikzf4 can directly convert mouse retinal glial cells into photoreceptor cells and reprogram mouse embryonic fibroblasts into neurons. These findings uncover the potential of temporal identity factors in reprogramming terminally differentiated cells.

Summary: Neural induction of human induced pluripotent stem cells involves the loss of pluripotency and the commitment to a neural lineage. This study analyzed the transcriptome of human iPSCs undergoing neural induction and identified functional modules involved in pluripotency loss, gain of neural identity, cell cycle, metabolism, cell fate commitment, genome integrity, stress response, and lineage specification. OTX2 was found to have a diverse role in neural induction and regulate processes such as protein remodeling, RNA splicing, and RNA processing. Inhibition of OTX2 accelerated pluripotency loss and disrupted neural induction.

Summary: The study found that hematopoietic stem cell (HSC) transplantation can repair radiation-induced lung tissue damage, with some HSCs potentially transdifferentiating into pneumonocytes. These data suggest that donor HSCs may migrate to the injured lung of the recipient, repairing the damage caused by radiation.

Summary: Recent research has shown that AMPK can significantly reduce alcohol-induced liver injury and enhance hepatocytes' mitophagy levels. AMPK indirectly upregulates the protein level of UQCRC2 by promoting NFE2L2/NRF2, thus improving mitophagy and alleviating liver damage.

Summary: Type 2 diabetes (T2D) is a disease caused by insulin resistance and insufficient insulin secretion. Pancreatic beta-cell dysfunction is the primary determinant of T2D progression and remission. Recent studies have shown that beta-cell dedifferentiation and reprogramming play important roles in the early and middle stages of T2D progression. Understanding the transcriptional control of beta-cell identity and plasticity can lead to the development of more effective strategies for treating T2D.

Summary: During liver organogenesis, a series of hepatic transcriptional regulators reshape cellular transcription profiles to activate hepatic genes. Pioneer factors open highly condensed chromatin to facilitate the action of other transcription factors. The bookmarking mechanism provides transcriptional competence for future activation of target genes, accompanied by a dynamic maturation process preparing for stable and efficient transcription.

Summary: Transcription factors bind to DNA in a sequence-specific manner and selectively impact gene expression, but their binding sites do not accurately predict the genes they directly control. A new study demonstrates that the same transcription factor binding sites have a greater impact on gene regulation during developmental change.

Summary: This study investigates the role of PDGF receptor a (PDGFRa) signaling in the differentiation of fibroblasts into myofibroblasts during skin wound healing. The results show that elevated PDGFRa signaling increases fibroblast proliferation but delays the fibroblast-to-myofibroblast transition. On the other hand, deletion of PDGFRa decreases cell proliferation and myofibroblast differentiation. Additionally, the coactivators MRTFA and MRTFB are found to be crucial for myofibroblast formation.

Summary: Temporal patterning is an important mechanism in generating diverse neuron subtypes from seemingly homogenous progenitor cells. By studying Drosophila optic lobe medulla, researchers discovered that two temporal transcription factors and cell-cycle dependent Notch signaling directly regulate the expression of TTFs Sloppy-paired 1/2 (Slp1/2) at the transcriptional level, revealing a novel Notch-pathway dependent mechanism in regulating the timing of a temporal transition within a TTF transcriptional cascade.

Summary: Type 1 diabetes is a chronic disease with autoimmune destruction of beta-cell population. Current treatments have significant side effects. Gene therapy and viral vectors have been explored for inducing pancreatic transdifferentiation to replace destroyed beta-cells. Studies using transcription factors have achieved varying levels of success, but further research and development are needed.

Summary: Transduction of the 4 transcription factors (Hnf4a, Foxa1, Prox1, and Hlf) promotes the differentiation of liver ductal organoids (LDOs) into mature hepatocyte-like cells with enhanced functionality, providing important resources for liver regenerative medicine and the study of physiological mechanisms.

Summary: In this article, a distributed hierarchical control strategy of thermostatically controlled loads (TCLs) is proposed to provide power dispatching service in a microgrid. The strategy includes a distributed consensus protocol to achieve optimal control performance and pinning control to distribute dispatching power among buildings. The case studies validate the effectiveness and fairness of the proposed control scheme, which also considers multiple factors of each TCL to improve the feasibility and expansibility of the dispatching process.

Summary: Renal imaging is widely used to assess surrogate markers of nephron mass, particularly in pediatric populations. Three-dimensional imaging technologies such as MRI and CT offer advantages over traditional ultrasound in accurately estimating kidney volume. Accounting for structural anomalies like hydronephrosis is crucial for estimating renal parenchymal area, as it correlates with chronic kidney disease progression.
