Summary: This study identified candidate genes involved in the biosynthesis of benzylisoquinoline alkaloids (BIAs) in Corydalis yanhusuo through full-length transcriptomic and metabolomic analyses. A complete BIA biosynthesis pathway was constructed based on the findings, and phylogenetic analysis revealed enzymes with columbamine-O-methyltransferase activity. The results provide a foundation for further molecular cloning of enzymes and activity validation studies.

Summary: Two pairs of novel polyphenolic compounds, named Yanhusuosines A and B, have been isolated from the tubers of Corydalis yanhusuo. Their molecular structures have been determined using spectroscopic and quantum-chemical calculation methods. These compounds represent a new class of alkaloid dimers biogenetically formed by a benzylisoquinoline molecule and a protoberberine unit through [4+3] cycloaddition. Compound 2 showed moderate inhibitory activity against NO formation in LPS-induced RAW264.7 macrophages.

Summary: This study evaluated the antiulcer effect of protopanaxatriol (PPT), a major intestinal bacterial metabolite of ginsenosides, in a rat model of gastric ulcer induced by acetic acid. High and medium doses of PPT significantly reduced ulcer area and regulated various serum markers in a dose-dependent manner. Metabolomics and network pharmacology analysis revealed key biomarkers, targets, and pathways involved in the antiulcer activity of PPT. The integrated analysis of metabolomics and network pharmacology provides an effective strategy for deciphering the mechanisms of natural compounds.
