We have previously identified 15 genes that are associated with the development of severe depressive side effects during the standard therapy with interferon alpha and ribavirin in the peripheral blood of hepatitis C virus infected patients. An enhanced expression of these genes was also found in the blood of psychiatric patients suffering severe depressive episode. Herein, we demonstrate that the same depression-related interferon-inducible genes (DRIIs) are also upregulated in postmortem brains of suicidal individuals. Using cultured mouse hippocampal and prefrontal neurons we show that costimulation with murine IFN (mIFN) and the TLR3 agonist poly(I: C) promotes the expression of the described DRIIs, at the same time inducing pro-inflammatory cytokine expression through Stat1 and Stat3 activation, promoting neuronal apoptosis. Consequently, the upregulation of selective DRIIs, production of inflammatory cytokines and inhibition of neuronal plasticity may be involved in the pathogenesis of IFN-associated depression.

Mesenchymal stromal cell-derived small extracellular vesicles promote neurological recovery and brain remodeling after distal middle cerebral artery occlusion in aged rats

Danut-Adrian Dumbrava, Roxana Surugiu, Verena Boerger, Mihai Ruscu, Tobias Tertel, Bernd Giebel, Dirk M. Hermann, Aurel Popa-Wagner

Summary: The study found that MSC-sEVs can improve neurological recovery and brain tissue remodeling in aged rats post-stroke by promoting angiogenesis and neurogenesis to alleviate the inflammatory environment after stroke.

Phosphorylation of PI3K/Akt at Thr308, but not phosphorylation of MAPK kinase, mediates lithium-induced neuroprotection against cerebral ischemia in mice

Summary: Lithium has neuroprotective effects after ischemic stroke, which involves modulation of the PI3K/Akt pathway but not the MEK/ERK pathway. High doses of lithium decrease brain perfusion and lead to animal death, while moderate doses improve brain tissue survival and reverse blood-brain barrier leakage, hemodynamic impairment, and apoptosis.

Tenascin-C restricts reactive astrogliosis in the ischemic brain

Summary: Cellular responses in glia play a key role in regulating brain remodeling post-stroke. Excessive glial reactivity impedes neuroplasticity and hampers neurological recovery. We found that tenascin-C (TnC) plays a central role in restraining astrogliosis and regulating astrocyte-microglial interactions.

Summary: CDNF and MANF, when administered after stroke, can promote neurological recovery and increase neuronal survival. They also reduce brain atrophy and glial scar formation. CDNF has the additional effect of increasing endogenous neurogenesis and promoting axonal growth.

Summary: The functions and activities of astroglia cells in brain development, aging, and neurodegenerative diseases differ. Comparing the transcriptomic activity of astroglia cells in these processes may provide new therapeutic strategies to protect the aging brain and improve clinical outcomes.

Structural changes in perineuronal nets and their perforating GABAergic synapses precede motor coordination recovery post stroke

Egor Dzyubenko, Katrin I. Willig, Dongpei Yin, Maryam Sardari, Erdin Tokmak, Patrick Labus, Ben Schmermund, Dirk M. Hermann

Summary: The study found that perineuronal nets (PNNs) undergo structural changes after ischemic stroke and these changes are associated with the recovery of motor coordination. The study also revealed that activated microglia/macrophages preferentially contact neurons coated with PNNs. These findings provide new insights into the mechanisms underlying neuroplasticity after stroke.

Summary: The evidence suggests that affective disorders increase the risk for cognitive decline and dementia in aging individuals. Cognitive decline is a central feature of aging and impacts quality of life. Motor deficits are common after brain injuries and stroke, affecting subjective well-being and quality of life.
