Summary: RO4929097 can effectively inhibit RANKL-induced osteoclastogenesis, reduce NFATc1 protein expression, and decrease bone resorption. In vivo experiments demonstrate that RO4929097 has an inhibitory effect on LPS-induced bone resorption.

Summary: The study evaluated the measurement of TRACP-5b and established reference ranges for a Western-European population, showing good concordance between the Nittobo and IDS methods. In CKD and HD patients, TRACP-5b and b-ALP were within their respective reference ranges, while beta-CTX increased with decreasing eGFR.

Summary: By studying the bone patterning during zebrafish fin ray regeneration, it was discovered that the fin rays form through two mineralization fronts and bifurcation is not a simple splitting process. Osteolytic tubules play a key role in defining the position of the branchpoint.

Summary: This study reveals that PTH signaling upregulates Notch pathway components, and Notch activation contributes to the catabolic actions of PTH in bone. Deleting the PTH receptor or overexpressing sclerostin in osteocytes abolishes the increase in Notch components caused by PTH. Moreover, bone-targeted Notch inhibition enhances the bone anabolic effects of intermittent PTH.

Summary: In this study, Babam2 was identified as an essential negative regulator of osteoclast formation and bone resorption. It interacts with Hey1 to inhibit Nfatc1 transcription, thus negatively regulating osteoclastogenesis. Targeting Babam2 may be a novel therapeutic approach for osteoclast-related bone diseases.

Summary: The study demonstrated the immunosuppressive function of IL-35 on RANKL and M-CSF-induced osteoclastogenesis and angiogenesis through Th17/IL-17 axis. Utilizing IL-35 as a therapeutic approach may serve as a novel treatment option for osteoporosis by suppressing bone resorption and angiogenesis.

Summary: This study showed that hesperidin impaired osteoclastogenesis in vitro, but high-dose dietary hesperidin increased osteoclast numbers and promoted inflammation-induced alveolar bone loss in vivo. However, hesperidin at a dose of 500 mg/kg exhibited a bone-sparing effect on skeletal bones under physiological conditions.

Summary: The Mir338 cluster is significantly enriched in gingival tissues with periodontitis and plays a role in both inflammation and alveolar bone loss. Knockout of the Mir338 cluster in mice attenuates alveolar bone loss and affects macrophage polarization and osteoclast formation. The Mir338 cluster could be a potential therapeutic target for periodontitis-related bone loss.

Summary: This study found that alpha-asarone (ASA), a compound derived from the traditional Chinese medicinal herb Acorus tatarinowii, can inhibit the activity of overactivated osteoclasts and reduce bone resorption. ASA's effects on osteoclastogenesis are dose-dependent and mediated through multiple signaling pathways. In an estrogen-deficiency-induced osteoporosis model, ASA also improved bone microstructure.

Summary: The study shows that uranium can have opposite effects on osteoclast resorption depending on its concentration in the bone microenvironment, and resorption can contribute to the remobilization of bone matrix-bound uranium. Through biomimetic matrices, the research highlights the complex mutual effects between osteoclasts and uranium, which helps to further study the impact of uranium on human bone health.

Summary: In this study, the functions of the OnTRAP5b enzyme in Oreochromis niloticus were investigated. The results showed that OnTRAP5b was most abundant in the liver and widely expressed in other tissues. Upon infection with Streptococcus agalactiae and Aeromonas hydrophila, the expression of OnTRAP5b was significantly up-regulated. The enzyme also exhibited optimal phosphatase activity at pH 5.0 and a temperature of 50 degrees C.
