Chronic stress is the major cause of clinical depression. The behavioral signs of depression, including anhedonia, learning and memory deficits, and sleep disruption, result from the damaging effects of stress hormones on specific neural pathways. The Chinese tree shrew (Tupaia belangeri chinensis) is an aggressive non-human primate with a hierarchical social structure that has become a well-established model of the behavioral, endocrine, and neurobiological changes associated with stress-induced depression. The tricyclic antidepressant clomipramine treats many of the core symptoms of depression in humans. To further test the validity of the tree shrew model of depression, we examined the effects of clomipramine on depression-like behaviors and physiological stress responses induced by social defeat in subordinate tree shrews. Social defeat led to weight loss, anhedonia (as measured by sucrose preference), unstable fluctuations in locomotor activity, sustained urinary cortisol elevation, irregular cortisol rhythms, and deficient hippocampal long-term potentiation (LTP). Clomipramine ameliorated anhedonia and irregular locomotor activity, and partially rescued the irregular cortisol rhythm. In contrast, weight loss increased, cortisol levels were even higher, and in vitro LTP was still impaired in the clomipramine treatment group. These results demonstrate the unique advantage of the tree shrew social defeat model of depression.

Oscillatory biomarkers of autism: evidence from the innate visual fear evoking paradigm

Chuanliang Han, Mingrou Guo, Xiaoyin Ke, Lanting Zeng, Meijia Li, Naem Haihambo, Jianping Lu, Liping Wang, Pengfei Wei

Summary: Autism spectrum disorder (ASD) is a neurodevelopmental disorder that has deficits in social and cognitive functioning. It is diagnosed subjectively, and objective criteria for early diagnosis are still being researched. A recent animal study found that the defensive response to looming stimuli was impaired in mice with ASD, but whether this effect exists in humans and can serve as a reliable clinical neural biomarker is uncertain. This study recorded electroencephalogram responses in children with ASD and typical developed children and found that alpha-band activity in the posterior brain region remained unchanged in the ASD group after looming stimuli, while it was suppressed in the TD group. This method could be a novel, objective way to detect ASD earlier.

Lifespan changes in cannabinoid 1 receptor mRNA expression in the female C57BL/6J mouse brain

Summary: Age-related characteristics in female brain functions show variations in different brain regions, and the expressions of certain genes are also influenced by age. The expression of CB1 receptor increases in some brain regions but decreases in others with age. These findings suggest the need for further investigation into age-related characteristics of female brain functions.

Protein phosphatase 2A deficiency in hippocampal CA1 inhibits priming effect of morphine on conditioned place preference in mice

Jing Dai, Ran Xie, Zhou-Na Sun, Xiao-Lin Kou, Jia-Qi Zhang, Cui Qi, Rui Liu, Xiang Gao, Jing Wang, Jun Gao

Summary: Studies have shown that protein phosphorylation is important in morphine abuse, but the role of protein phosphatase 2A (PP2A) in the morphine-priming process is unclear. In this study, conditional knockout mice were used to investigate the role of hippocampal PP2A in morphine priming. The results suggest that hippocampal PP2A may be involved in morphine priming through the PP2A/HDAC4/Rack1 pathway.

Feasibility of using AI to auto-catch responsible frames in ultrasound screening for breast cancer diagnosis

Jing Chen, Yitao Jiang, Keen Yang, Xiuqin Ye, Chen Cui, Siyuan Shi, Huaiyu Wu, Hongtian Tian, Di Song, Jincao Yao, Liping Wang, Sijing Huang, Jinfeng Xu, Dong Xu, Fajin Dong

Summary: This research explores the method of using artificial intelligence to capture complementary responsible frames from breast ultrasound screening in AI-assisted breast diagnosis. The AI model based on FEBrNet-recommended frames outperforms other frame set based AI models, as well as ultrasound and mammography physicians, indicating the level of FEBrNet reaching medical specialists.

Hydroxychloroquine protects against autoimmune premature ovarian insufficiency by modulating the Treg/Th17 cell ratio in BALB/c mice

Ling Shuai, Jiajie She, Ruiying Diao, Huihui Zhao, Xuemin Liu, Qingyu Hu, Dongdong Li, Danna Su, Xiaofeng Ye, Yan Guo, Mei Zhong, Liping Wang

Summary: This study investigated the effect of hydroxychloroquine (HCQ) on ovarian function in mice with premature ovarian insufficiency (POI). The results showed that HCQ restored ovarian function, increased estradiol levels, and decreased follicle stimulating hormone levels. HCQ also inhibited the activation of the TLR7 signaling pathway, preventing inflammation and the differentiation of CD4(+) T cells into Th17 cells. These findings suggest that HCQ may be a promising therapeutic method for patients with POI.

Systematic evaluation of a predator stress model of depression in mice using a hierarchical 3D-motion learning framework

Yu-Ting Tseng, Binghao Zhao, Hui Ding, Lisha Liang, Bernhard Schaefke, Liping Wang

Summary: Investigation into neurobiology of depression requires animal models that mimic specific aspects of the human disorder. However, commonly used paradigms have gender biases when applied to female mice. This study shows that predator stress effectively induces depression-like behaviors in both male and female mice, and machine learning-based analysis can predict depression status from behavioral patterns. The study highlights the importance of unbiased evaluation and provides a holistic approach for studying neuropsychiatric disorders.

A morphine reward generalization mouse model based on conditioned place preference and aversion

Summary: In this study, a simple and effective experimental model was designed to investigate the generalization memory of morphine reward. By replacing the conditioning CPP box, it was found that mice still showed a clear preference for generalized information. The establishment of this study provides a new tool for studying generalization memory in substance use disorders.

Altered pupil responses to social and non-social stimuli in Shank3 mutant dogs

Wei Ren, Kang Huang, Yumo Li, Qin Yang, Liping Wang, Kun Guo, Pengfei Wei, Yong Q. Zhang

Summary: This study developed a pupilometer for dogs and used it to measure real-time pupillary responses. The researchers found that gene-edited dogs carrying mutations in Shank3 showed altered pupillary responses to light, loud noise, and high reward food. The Shank3 mutant dogs also exhibited impaired pupillary synchrony during dog-human interaction. These findings suggest that the pupilometer can be used to study the underlying neural mechanisms of autism spectrum disorder and other psychiatric disorders.

Defensive responses: behaviour, the brain and the body

Yu-Ting Tseng, Bernhard Schaefke, Pengfei Wei, Liping Wang

Summary: Animals need to balance defensive responses against predators with other adaptive behaviors. Accumulating evidence suggests that endocrine, immune, gastrointestinal, and reproductive systems play important roles in modulating behavioral responses to threat. Understanding the interconnected network between the brain and body is crucial for appropriate defensive responses to threats.
