Use of Recombinant Tobacco Mosaic Virus To Achieve RNA Interference in Plants against the Citrus Mealybug, Planococcus citri (Hemiptera: Pseudococcidae)

The citrus mealybug, Planococcus citri, is an important plant pest with a very broad plant host range. P. citri is a phloem feeder and loss of plant vigor and stunting are characteristic symptoms induced on a range of host plants, but P. citri also reduces fruit quality and causes fruit drop leading to significant yield reductions. Better strategies for managing this pest are greatly needed. RNA interference (RNAi) is an emerging tool for functional genomics studies and is being investigated as a practical tool for highly targeted insect control. Here we investigated whether RNAi effects can be induced in P. citri and whether candidate mRNAs could be identified as possible targets for RNAi-based P. citri control. RNAi effects were induced in P. citri, as demonstrated by specific target reductions of P. citri actin, chitin synthase 1 and V-ATPase mRNAs after injection of the corresponding specific double-stranded RNA inducers. We also used recombinant Tobacco mosaic virus (TMV) to express these RNAi effectors in Nicotiana benthamiana plants. We found that P. citri showed lower fecundity and pronounced death of crawlers after feeding on recombinant TMV-infected plants. Taken together, our data show that actin, chitin synthase 1 and V-ATPase mRNAs are potential targets for RNAi against P. citri, and that recombinant TMV is an effective tool for evaluating candidate RNAi effectors in plants.

Biofabrication of ZnO nanoparticles using Acacia arabica leaf extract and their antibiofilm and antioxidant potential against foodborne pathogens

Summary: In this study, zinc oxide nanoparticles (ZnO NPs) were synthesized from aqueous leaf extract of Acacia arabica and characterized. The results revealed that the green synthesized ZnO NPs exhibited strong antibacterial activity against foodborne pathogens, inhibiting both biofilm formation and exopolysaccharide production. The nanoparticles also showed significant antioxidant potential and were non-toxic to HeLa cell lines at the tested concentrations. These findings suggest that green synthesized ZnO NPs can be used as alternative therapeutic agents against antibiotic resistant foodborne pathogens.

A DNA barcode survey of insect biodiversity in Pakistan

Summary: This study used DNA barcoding to survey the insect diversity in Pakistan and found that the knowledge about insect species in Pakistan is limited. Most species can only be classified into order and family, with only a small portion classified into genus and species. The study also found that a majority of the insect species in Pakistan are unique to the country, but some species are also reported in neighboring countries.

Ecofriendly phytosynthesized zirconium oxide nanoparticles as antibiofilm and quorum quenching agents against Acinetobacter baumannii

Summary: This study synthesized zirconium oxide nanoparticles (ZrO2 NPs) from aqueous ginger extract and evaluated their antibiofilm and quorum quenching (QQ) potential against multi-drug resistant (MDR) Acinetobacter baumannii. The results demonstrated that ZrO2 NPs could generate reactive oxygen species (ROS) to disrupt the bacterial cellular membrane and inhibit biofilm formation by altering its chemical composition. Molecular docking studies also revealed the interaction between ZrO2 NPs and proteins involved in biofilm formation. Overall, the phytosynthesized ZrO2 NPs showed promising efficacy as alternative therapeutic options against MDR A. baumannii.

Plant extracts rich in polyphenols can be an effective agent for wound healing, but optimization of the extraction protocol is needed to ensure efficacy.

Behavioural impairments, foraging behaviour and brood development of Apis mellifera L. (Hymenoptera: Apidae) driven by air pollutants particulate matter in agro-industrial ecosystem

Summary: Honey bees are effective sentinels for environmental contaminants, particularly particulate matter. Exposure to particulate matter had harmful effects on the foraging performances of honey bees, resulting in reduced foraging rates and speeds. The presence of particulate matter also led to a decrease in bee population, smaller broods, and lesser honey storage. Additionally, the presence of particulate matter affected the olfaction and flight performance of honey bees. The study also found an increase in sodium and potassium levels in the body fluids of bees exposed to agro-industrial particulate matter. The findings emphasize the importance of preventing the negative effects of particulate matter on bees' behavior and development.

Native and Non-Native Bemisia tabaci NAFME Haplotypes Can Be Implicated in Dispersal of Endemic and Introduced Begomoviruses in Oman

Summary: The prevalence and relationships between putative native and exotic begomoviruses and the North Africa-Middle East haplotypes of B. tabaci were investigated in Oman. The study found close associations between various begomoviral species and the B. tabaci insect pest in the region. The results suggest that irrigated agriculture and global trade have facilitated the diversification and spread of begomoviruses.
