Summary: Delta opioids have tissue-protective properties and may relieve ischemic injury through transactivation of EGFR. Pre-treatment with a delta opioid receptor agonist improved neurologic deficits and reduced infarct volume in a rat model of ischemic stroke, effects that were blocked by EGFR and MMP inhibitors. Activation of EGFR, Akt, and ERK1/2 and upregulation of EGFR expression by the delta opioid agonist were inhibited by AG1478 and GM6001, suggesting an essential role of MMP-mediated transactivation of EGFR in the neuroprotective effects of delta opioids.
