Aging affects mitochondria in a tissue-specific manner. Calorie restriction (CR) is, so far, the only intervention able to delay or prevent the onset of several age-related changes also in mitochondria. Using livers from middle age (18-month-old), 28-month-old and 32-month-old ad libitum-fed and 28-month-old calorie-restricted rats we found an age-related decrease in mitochondrial DNA (mtDNA) content and mitochondrial transcription factor A (TFAM) amount, fully prevented by CR. We revealed also an age-related decrease, completely prevented by CR, for the proteins PGC-1 alpha NRF-1 and cytochrome c oxidase subunit IV, supporting the efficiency of CR to forestall the age-related decrease in mitochondrial biogenesis. Furthermore, CR counteracted the age-related increase in oxidative damage to proteins, represented by the increased amount of oxidized peroxiredoxins (PRX-SO3) in the ad libitum-fed animals. An unexpected age-related decrease in the mitochondrial proteins peroxiredoxin III (Prx III) and superoxide dismutase 2 (SOD2), usually induced by increased ROS and involved in mitochondrial biogenesis, suggested a prevailing relevance of the age-reduced mitochondrial biogenesis above the induction by ROS in the regulation of expression of these genes with aging. The partial prevention of the decrease in Prx III and SOD2 proteins by CR also supported the preservation of mitochondrial biogenesis in the anti-aging action of CR. To investigate further the age- and CR-related effects on mitochondrial biogenesis we analyzed the in vivo binding of TFAM to specific mtDNA regions and demonstrated a marked increase in the TFAM-bound amounts of mtDNA at both origins of replication with aging, fully prevented by CR. A novel, positive correlation between the paired amounts of TFAM-bound mtDNA at these sub-regions was found in the joined middle age ad libitum-fed and 28-month-old calorie-restricted groups, but not in the 28-month-old ad libitum-fed counterpart suggesting a quite different modulation of TFAM binding at both origins of replication in aging and CR.

Targeting mitochondrial impairment for the treatment of cardiovascular diseases: From hypertension to ischemia-reperfusion injury, searching for new pharmacological targets

Simona Todisco, Biagia Musio, Vito Pesce, Maria Maddalena Cavalluzzi, Giuseppe Petrosillo, Gianluigi La Piana, Maria Noemi Sgobba, Nikola Schlosserova, Lucas Cafferati Beltrame, Rosa Di Lorenzo, Vincenzo Tragni, Domenico Marzulli, Lorenzo Guerra, Anna De Grassi, Vito Gallo, Mariateresa Volpicella, Luigi Leonardo Palese, Giovanni Lentini, Ciro Leonardo Pierri

Summary: Mitochondria and mitochondrial proteins are promising pharmacological target candidates for the prevention of hypertension and cardiovascular diseases. Targeting mitochondrial dysfunction in cardiomyocytes can prevent heart failure. Advances in omics approaches and computational methods allow for detailed investigation of impaired mitochondrial pathways in CVDs and the design of new selective drugs.

Summary: Altered mitochondrial quality in muscle may contribute to age-related decline in physical function. This study investigated the relationship between autophagy, mitophagy, lysosomal markers and measures of physical performance and lower extremity tissue composition in young and older adults. The results revealed that older adults had smaller muscle volume, lower tissue composition index, and higher protein content of p62 and BNIP3. The expression levels of p62 and BNIP3 were negatively correlated with the tissue composition index and performance on the 5-time sit-to-stand test. These findings suggest that dysfunction in the autophagy/mitophagy-lysosomal system is associated with deterioration of lower extremity tissue composition and muscle dysfunction.

Summary: This study aimed to investigate the effects of low muscle mass (LMM) on the risk of all-cause and cardiovascular disease (CVD)-specific mortality in cancer survivors. The results showed that LMM was positively associated with the risk of all-cause and CVD-specific mortality in cancer survivors.

High Protein Intake at Lunch Is Negatively Associated with Blood Pressure in Community-Dwelling Older Adults: A Cross-Sectional Study

Summary: The present study found that protein intake at lunch was independently and negatively associated with systolic blood pressure in community-dwelling older adults in Brazil. Additionally, participants with higher protein intakes had a lower prevalence of hypertension diagnosed by a physician. These results remained significant after adjusting for several covariates but lost significance when kilocalories and micronutrients were included in the model.

Inadequate Physical Activity Is Associated with Worse Physical Function in a Sample of COVID-19 Survivors with Post-Acute Symptoms

Summary: This study aimed to evaluate the association between physical activity levels and parameters of physical function in COVID-19 survivors with post-acute symptoms. The results showed that stopping physical activity after COVID-19 results in comparable physical performance to those who were never active. However, older adults who continue to engage in physical activity perform better in the six-minute walking test than sedentary peers.

Mitochondrial-Derived Vesicles: The Good, the Bad, and the Ugly

Anna Picca, Flora Guerra, Riccardo Calvani, Helio Jose Coelho, Francesco Landi, Cecilia Bucci, Emanuele Marzetti

Summary: Mitophagy plays a crucial role in maintaining mitochondrial quality, and extracellular vesicles (EVs) containing mitochondrial components are released through the endosomal-lysosomal system. This process occurs in response to mild mitochondrial damage and lysosomal dysfunction, with mitochondrial-derived vesicles (MDVs) helping to limit the release of proinflammatory mitochondrial damage-associated molecular patterns.
