Mounting evidence has indicated that the cardiovascular protective effects of dietary alpha-linolenic acid (ALA), but whether ALA exerts an endothelial protective effect against high glucose injury and the underlying mechanisms remain largely unknown. Streptozocin-induced diabetic rats were randomized treated orally for 4 weeks with vehicle (0.01% alcohol) or ALA (500 mu g/kg per day by gavage). Human umbilical vein endothelial cells (HUVECs) were exposed to high glucose (28 mmol/L) stimulation for 48 hours. ALA significantly improved concentration-dependent vasorelaxation to ACh in diabetic aortic segments and inhibited endothelial inflammation as evidenced by decreased soluble P-selectin and intercellular adhesion molecule-1 (ICAM-1) in diabetic rats. Furthermore, both P-selectin and ICAM-1 expression were increased significantly in high glucose-induced HUVECs, resulting in enhanced neutrophils adhesion to HUVECs compared with normal glucose group. Treatment with ALA (50 mmol/L) increased Akt phosphorylation, attenuated P-selectin and ICAM-1 expressions and thus inhibited neutrophils adhesion in HUVECs exposed to high glucose, all of which was blocked by the PI3K inhibitors LY294002 and wortmannin. These data indicates that ALA inhibits endothelial inflammation and improved endothelial function in STZ-induced diabetic rats. The anti-adhesive effect of ALA against high glucose injury may partially be mediated by the PI3K/Akt pathway.

Upregulated hepatokine fetuin B aggravates myocardial ischemia/reperfusion injury through inhibiting insulin signaling in diabetic mice

Wenjuan Xing, Yanzhen Tan, Kaifeng Li, Pei Tian, Fei Tian, Haifeng Zhang

Summary: In patients with type 2 diabetes mellitus, high levels of hepatokine fetuin-B exacerbate myocardial ischemia/reperfusion injury by interfering with insulin signaling pathways, leading to increased apoptosis and cardiac dysfunction. Conversely, knockdown of FetB can alleviate MI/R injury and improve insulin-induced signaling in diabetic hearts.

Summary: The study demonstrated that FYC can significantly alleviate the pathological damage and inflammatory cell infiltration in the colon tissue of UC mice induced by dextran sulfate sodium, restore colon length, reduce DAI score, MPO content, pro-inflammatory factors, and increase anti-inflammatory factors. The protective effect of FYC on UC is achieved by inhibiting the OSM/OSMR pathway and regulating inflammatory factors to improve the intestinal flora.

Summary: The study demonstrated that a single exposure to mild or severe hypobaric hypoxia could improve performance in healthy mice. In the cerebellum, 6 out of 8 detected genes showed no change after SHH, while in the hippocampus all genes remained unchanged. In the muscles, 7 out of 8 detected genes did not show any changes after SHH exposure.

Summary: This study demonstrated that TSG could improve endothelial function and reduce obesity-related hypertension by increasing omentin-1 levels, activating Akt/eNOS signaling pathway, and attenuating oxidative/nitrative stress. The down-regulation of omentin-1 is associated with endothelial dysfunction and hypertension in obesity, highlighting the potential of TSG as an antihypertensive agent for cardiovascular protection in obesity.
