Summary: Contrary to prevailing beliefs, angiogenesis in ischemic muscle injury is driven by the formation of wide-caliber conduits that rapidly transform into a hierarchical neocirculation. Inhibiting VEGFR2 accelerates this process, and endothelial cells in low-flow lumens display enhanced VEGFR2 mechanosignals.

Summary: Due to the complex mechanism and limited treatments available, the development of targeted drugs or inhibitors based on molecular mechanisms remains an important strategy for prevention and treatment. This paper describes the downstream signaling pathways mediated by VEGFR and LPAR1 in pulmonary cells, their role in pulmonary fibrosis, and the current status of drug research on LPAR1 and VEGFR2 targets. It analyzes the mechanisms by which these pathways regulate vascular leakage and collagen deposition leading to the development of pulmonary fibrosis, and discusses the mutual promotion of the two pathways. The development of drugs targeting both LPAR1 and VEGFR2 is proposed, along with important considerations in targeting and safety.

Summary: In this study, researchers investigated the role of VEGFR2 monoclonal antibodies in inhibiting tumorigenicity and VM formation in GBM. They found that VEGFR2 is highly expressed in glioma tissues and is associated with poor prognosis and VM markers. The monoclonal antibodies could inhibit invasion, migration, and VM formation in vitro and also inhibit tumor growth and VM formation in vivo. This suggests that VEGFR2 monoclonal antibodies could be a potential supplementary treatment for GBM.

Summary: Lung cancer is a common and deadly form of cancer, with non-small cell lung cancer (NSCLC) being the most diagnosed subtype. The VEGFR2 protein, a member of the VEGF receptor tyrosine kinase family, and Musashi-2 (MSI2) RNA-binding protein play important roles in NSCLC progression. This study demonstrates that MSI2 positively regulates VEGFR2 protein and impacts AKT signaling through negative regulation of PTEN mRNA translation. MSI2 directly binds to both VEGFR2 and PTEN mRNAs. Additionally, MSI2 expression correlates with VEGFR2 and VEGF-A protein levels in human lung adenocarcinoma samples. These findings suggest that the MSI2/VEGFR2 axis is involved in lung adenocarcinoma progression and should be further investigated as a potential therapeutic target.

Summary: This study investigated the impact of connexin 43 (Cx43) on corneal neovascularization and its regulation of VEGFR2 on vascular endothelial cells. The findings revealed that Cx43 regulates angiogenesis through the β-catenin-VE-cadherin-VEGFR2-Erk pathway. Gap26 was found to downregulate VEGFR2 phosphorylation by stabilizing the expression of β-catenin and VE-cadherin on the cell membrane, resulting in the inhibition of VEGFA-induced HUVECs proliferation, migration, tube formation, and corneal neovascularization.

Summary: This study aimed to evaluate the antifibrotic activity of sclareol (SCL) from Salvia sclarea on liver fibrosis and explore its underlying mechanisms. The results showed that SCL treatment inhibited the profibrotic tendency of activated hepatic stellate cells. In animal models, SCL administration reduced liver injury and collagen accumulation. Mechanistic studies revealed that SCL mediated VEGFR2 SUMOylation, resulting in the inhibition of the interaction between VEGFR2 and STAT3 and the suppression of downstream STAT3 phosphorylation.

Summary: Jujuboside B (JuB), a triterpenoid saponin from Ziziphus jujuba, was found to inhibit the proliferation, migration, and tube formation of human umbilical vein endothelial cells in a dose-dependent manner. It also suppressed angiogenesis and delayed the growth of colorectal cancer xenograft in mice. The molecular mechanism involved the inhibition of VEGFR2 phosphorylation and downstream signaling pathways. JuB has potential as a drug candidate for antiangiogenic cancer therapy.
