Protein synthesis is mainly regulated at the initiation step, allowing the fast, reversible and spatial control of gene expression. Initiation of protein synthesis requires at least 13 translation initiation factors to assemble the 80S ribosomal initiation complex. Loss of translation control may result in cell malignant transformation. Here, we asked whether translational initiation factors could be regulated by NF-kappa B transcription factor, a major regulator of genes involved in cell proliferation, survival, and inflammatory response. We show that the p65 subunit of NF-kB activates the transcription of eIF4H gene, which is the regulatory subunit of eIF4A, the most relevant RNA helicase in translation initiation. The p65-dependent transcriptional activation of eIF4H increased the eIF4H protein content augmenting the rate of global protein synthesis. In this context, our results provide novel insights into protein synthesis regulation in response to NF-kB activation signalling, suggesting a transcription-translation coupled mechanism of control.

Summary: This study investigates the capacity of the rare disease healthcare network in Campania to diagnose patients with rare diseases during the Covid-19 outbreak, and sheds light on problematic diagnoses during this period. The results show that the prevalence of new diagnoses of rare disease was significantly lower during the outbreak, and various issues such as lack of diagnostic resources and guidelines were identified.

IBtkα Activates the β-Catenin-Dependent Transcription of MYC through Ubiquitylation and Proteasomal Degradation of GSK3β in Cancerous B Cells

Summary: The IBTK gene encodes the IBtk alpha protein, which plays a role in the regulation of MYC-dependent B-lymphomagenesis. Mechanistically, IBtk alpha indirectly activates the beta-catenin-dependent transcription of the MYC gene by modulating GSK3 beta. Silencing IBtk alpha leads to downregulation of MYC expression and increased apoptosis in Burkitt's lymphoma cells.

Summary: Chronic Lymphocytic Leukemia (CLL) is a heterogeneous disease with variable clinical courses among different patients. This study discovered the possible existence of multiple independent CLL clones within the same patient. By selecting a peptide binder that binds to specific clone cells using phage display libraries and performing flow cytometry, it was found that the specific clone cells expressed more genes involved in survival and apoptosis escape processes compared to other clone cells, and they did not carry characteristic genetic lesions.

Summary: Breast cancer-associated fibroblasts (BCAFs) promote breast cancer progression by interacting with cancer cells. In an in vitro experiment, BCAFs became inactivated after 216 hours of 3D culture, resulting in a cytostatic effect on MCF-7 cells. Reverted BCAFs also exhibited an inactivated phenotype and inhibited the migration of MCF-7 cells. These findings suggest that deactivation of BCAFs without drug treatment reduces their ability to sustain breast cancer progression in vitro.

Preliminary data indicated that dupilumab treatment led to a reduction in inflammation and modulation of specific miRNAs related to proliferation, inflammation, and drug-resistance.

Severe and mild-moderate SARS-CoV-2 vaccinated patients show different frequencies of IFNγ-releasing cells: An exploratory study

Summary: In this study, we evaluated the spike-specific immune response in patients with breakthrough SARS-CoV-2 infection after receiving two doses of the BNT162b2 mRNA vaccine. The results showed that severe COVID-19 patients requiring hospitalization had lower levels of spike-specific IFN gamma-releasing cells compared to mild-moderate patients and healthy subjects, despite similar antibody titers.

Development of Cyclic Peptides Targeting the Epidermal Growth Factor Receptor in Mesenchymal Triple-Negative Breast Cancer Subtype

Summary: In this study, we used phage display to select two highly specific peptide ligands for targeting the overexpressed EGFR in TNBC cells. Molecular docking predicted the peptides' binding affinities and sites on the extracellular domain of EGFR. Flow cytometry validated the binding of FITC-conjugated peptides to TNBC cells. Confocal microscopy confirmed the peptide binding specificity to EGFR-positive tumor tissues. These peptides could be used in conjunction with nanoparticles for tumor-targeted delivery of anticancer drugs.

Can early physical therapy positively affect the onset of independent walking in infants with Down syndrome? A retrospective cohort study
