Summary: An INDIRECT active targeting strategy, called INTACT, is proposed to address the problem of on-target toxicity in antibody-based therapies. By prebinding antibodies with cell membrane vesicles that have appropriate receptor expressions, this strategy filters out interactions on nontarget sites while ensuring release at the targets by competitive binding. The strategy is effective in alleviating hematotoxicity and achieving therapeutic effects in animal models, and it has broad applicability for various systems, providing a potential platform for improving the specificity of antibody-related therapies.

Summary: The nanoscale geometry of cell-supported lipid bilayers (SLBs) strongly affects the binding of cell adhesion receptors to ligands, as the adhering membrane bends to conform to the SLB geometry for receptor-ligand binding. This finding may have profound implications for the future development of tissue engineering and regenerative medicine.

Summary: A self-assembled magnetic micelle delivery system was developed for targeted delivery of CD8(+) T-cell epitopes, producing strong cellular immune responses. Metal ions enhanced vaccine immunogenicity, while a self-immolative linker allowed for rapid antigen release. The system eradicated tumors in a mouse model, demonstrating potential for developing a new generation of cancer vaccines with multiple functionalities.

Summary: Many pathogenic bacteria are becoming more resistant to antibiotics, especially in the form of biofilms. Photodynamic therapy, which uses reactive oxygen species (ROS) produced by a photosensitizer, is a promising alternative method for treating microbial infections. However, the unspecific activity of ROS can also harm healthy tissue. This study focuses on mesoporous organosilica colloids functionalized by click-chemistry methods to target and detect biofilms, with successful efficacy demonstrated on Pseudomonas fluorescens biofilms.

Summary: In this study, pathological collagen targeting and penetrating liposomes were developed to deliver dual anti-fibrotic drugs deep into the injured alveoli of patients with IPF. The liposomes were decorated with collagen binding peptide and collagenase, allowing them to recognize and target the pathological collagen efficiently. The released dual drugs exhibited a synergistic anti-fibrotic effect, repairing the damaged epithelium and remodeling the extracellular matrix to rebuild the lung architecture.

Summary: Lymphocyte activation gene 3 protein (LAG3) is an inhibitory receptor highly expressed on exhausted T cells in tumors and chronic viral infection, making it a major immunotherapeutic target. By studying seven monoclonal antibodies that recognize LAG3, it was found that they mainly target the first Ig-like domain (D1) of LAG3 and partially overlap with the binding site for MHC class II.

Summary: Abnormal glycosylation is associated with cancer cell trafficking and drug resistance. This study reveals that desialylation enhances NK cell-mediated cytotoxicity against multiple myeloma (MM) cells and increases detection of CD38. These findings suggest that desialylation could enhance the anti-MM response of frontline therapies.
