Urinary ATP May Be a Dynamic Biomarker of Detrusor Overactivity in Women with Overactive Bladder Syndrome

Background: Nowadays, there is a considerable bulk of evidence showing that ATP has a prominent role in the regulation of human urinary bladder function and in the pathophysiology of detrusor overactivity. ATP mediates nonadrenergic-noncholinergic detrusor contractions in overactive bladders. In vitro studies have demonstrated that uroepithelial cells and cholinergic nerves from overactive human bladder samples (OAB) release more ATP than controls. Here, we compared the urinary ATP concentration in samples collected non-invasively from OAB women with detrusor overactivity and age-matched controls. Methods: Patients with neurologic diseases, history of malignancy, urinary tract infections or renal impairment (creatinine clearance <70 ml/min) were excluded. All patients completed a 3-day voiding diary, a 24 h urine collection and blood sampling to evaluate creatinine clearance. Urine samples collected during voluntary voids were immediately freeze-preserved for ATP determination by the luciferin-luciferase bioluminescence assay; for comparison purposes, samples were also tested for urinary nerve growth factor (NGF) by ELISA. Results: The urinary content of ATP, but not of NGF, normalized to patients' urine creatinine levels (ATP/Cr) or urinary volume (ATP. Vol) were significantly (P<0.05) higher in OAB women with detrusor overactivity (n = 34) than in healthy controls (n = 30). Significant differences between the two groups were still observed by boosting urinary ATP/Cr content after water intake, but these were not detected for NGF/Cr. In OAB patients, urinary ATP/Cr levels correlated inversely with mean voided volumes determined in a 3-day voiding diary. Conclusion: A high area under the receiver operator characteristics (ROC) curve (0.741; 95% CI 0.62-0.86; P<0.001) is consistent with urinary ATP/Cr being a highly sensitive dynamic biomarker for assessing detrusor overactivity in women with OAB syndrome.

Could hypoxia rehabilitate the osteochondral diseased interface? Lessons from the interplay of hypoxia and purinergic signals elsewhere

Summary: The osteochondral unit consists of articular cartilage, subchondral bone, and calcified cartilage. Cells within this unit, such as chondrocytes and osteoblasts, release nucleotides to the local environment, which plays a role in tissue homeostasis. Hypoxic conditions can affect the expression and activity of purinergic signaling molecules, and the interplay between hypoxia and the purinergic signaling cascade contributes to osteochondral unit homeostasis. Understanding this relationship may lead to new therapeutic targets for osteochondral rehabilitation.

Summary: The anti-obesity hormones T3 and irisin have been found to stimulate thermogenesis and energy consumption, while reducing lipid accumulation. This study investigated their effects on the purinergic signaling pathway in adipocytes, and found that T3 and irisin differentially regulate the transcription of purinergic signaling markers. These findings may offer new insights into the treatment of obesity and related metabolic disorders.

Purinergic Tuning of the Tripartite Neuromuscular Synapse

Summary: The vertebrate neuromuscular junction is a specialised synapse composed of a presynaptic region, a postsynaptic skeletal motor endplate area, and perisynaptic Schwann cells that play a significant role in maintaining neuromuscular integrity, transmission, and plasticity. Acetylcholine is the main neurotransmitter at this synapse and is modulated by purinergic neuromodulators. Adenine nucleotides and adenosine, released alongside acetylcholine, regulate transmitter release and expression of ACh receptors via P2Y and P2X receptors. Adenosine, through A1 and A2A receptors, also modulates ACh release by motor nerve terminals. Understanding the complex interplay between purines and neuromuscular transmission is essential for conditions like Myasthenia gravis.

Age-related white matter hyperintensities and overactive bladder: A systematic review

Using Raw Accelerometer Data to Predict High-Impact Mechanical Loading

Summary: The purpose of this study was to develop prediction equations for peak ground reaction force (pGRF) and peak loading rate (pLR) in high-impact activities for adults with various body masses. Regression equations were developed using accelerometry data, with body mass and peak acceleration or acceleration rate as predictors. The equations had high accuracy for predicting pGRF, with a coefficient of determination (R-2) of at least 0.83 and a mean absolute percentage error (MAPE) below 14.5%. However, the accuracy of the pLR prediction equations was lower than that of the pGRF equations.

Renal cell carcinoma in native kidneys before transplantation-When will we stop waiting?

Summary: Kidney transplantation has little impact on the tumor progression risk of renal cell carcinoma. Kidney transplant recipients have similar tumor recurrence and overall survival rates as dialyzed patients. The only factor associated with worse survival is the duration of dialysis.

External Validation of the Toulouse-Rangueil Predictive Model to Estimate Donor Renal Function After Living Donor Nephrectomy

Retrospective analysis of living donor kidney transplants from 1998 to 2020 revealed a good agreement between predicted and observed eGFR at 1-year post-donation. The model demonstrated a good discriminative ability in predicting CKD at 1-year post-donation.

Maximal Exercise Improves the Levels of Endothelial Progenitor Cells in Heart Failure Patients

Summary: This study aims to evaluate the effects of a single exercise bout on the circulating levels of EPCs and CECs in heart failure patients. The results showed that a single exercise bout increased the levels of EPCs but didn't change the levels of CECs. Heart failure patients had lower levels of EPCs compared to the age-matched group, but the exercise bout restored their EPCs levels to a level similar to the age-matched group.

Summary: This study aimed to investigate the involvement of the P2X7 receptor blocker Brilliant Blue G (BBG) in gastric emptying and oxidative stress changes in the gastrointestinal tract of spontaneously hypertensive rats (SHR), and to study its relationship with the renin-angiotensin system. The results showed that BBG improved gastric emptying delay and oxidative stress markers in SHR animals, and this preventive effect was abrogated by the renin-angiotensin system.
