KLF5 is a basic transcription factor that regulates multiple biological processes. While it was identified as a putative tumor suppressor in prostate cancer, likely due to its function as an effector of TGF-beta in the inhibition of cell proliferation, KLF5 is unacetylated and promotes cell proliferation in the absence of TGF-beta. In this study, we evaluated the expression and function of KLF5 in prostatic epithelial homeostasis and tumorigenesis using mouse prostates and human prostate epithelial cells in 3-D culture. Histological and molecular analyses demonstrated that unacetylated-Klf5 was expressed in basal or undifferentiated cells, whereas acetylated-Klf5 was expressed primarily in luminal and/or differentiated cells. Androgen depletion via castration increased both the level of Klf5 expression and the number of Klf5-positive cells in the remaining prostate. Functionally, knockdown of KLF5 in the human RWPE-1 prostate cell line decreased the number of spheres formed in 3-D culture. In addition, knockout of Klf5 in prostate epithelial cells, mediated by probasin promoter-driven Cre expression, did not cause neoplasia but promoted cell proliferation and induced hyperplasia when one Klf5 allele was knocked out. Knockout of both Klf5 alleles however, caused apoptosis rather than cell proliferation in the epithelium. In castrated mice, knockout of Klf5 resulted in more severe shrinkage of the prostate. These results suggest that KLF5 plays a role in the proliferation and differentiation of prostatic epithelial cells, yet loss of KLF5 alone is insufficient to induce malignant transformation in epithelial cells.

Summary: In renal cell carcinoma (RCC), the interaction between long non-coding RNA (lncRNA) HOTAIR and androgen receptor (AR) forms a feedback loop that promotes tumor angiogenesis, with HOTAIR enhancing GLI2 expression by cooperatively binding to its promoter with AR. These findings suggest that HOTAIR and GLI2 could serve as novel therapeutic targets against RCC.

Novel Gene Signatures Predictive of Patient Recurrence-Free Survival and Castration Resistance in Prostate Cancer

Summary: Molecular signatures predictive of recurrence-free survival and castration resistance in prostate cancer are crucial for treatment decisions, but current signatures have limited robustness. By analyzing 287 differentially expressed genes, a 6-gene signature called CRPC-derived prognosis signature (CRPCPS) was identified to predict recurrence-free survival and distinguish between castration-resistant and hormone-sensitive prostate cancer. The robustness of this signature was demonstrated in multiple cohorts, showing its potential clinical utility.

The Cardiac Glycoside Deslanoside Exerts Anticancer Activity in Prostate Cancer Cells by Modulating Multiple Signaling Pathways

Mingcheng Liu, Qingqing Huang, A. Jun, Linyue Li, Xiawei Li, Zhiqian Zhang, Jin-Tang Dong

Summary: Prostate cancer is a major cause of cancer-related deaths among men, requiring novel treatments. Deslanoside has shown anticancer activity in prostate cancer cells by modulating gene expression, inducing cell cycle arrest at G2/M, and promoting apoptosis. The altered expression of specific genes by deslanoside also correlates with survival outcomes in prostate cancer patients, suggesting potential for its repurposing as an anticancer drug.

A novel PDT: 5-aminolevulinic acid combined 450 nm blue laser photodynamic therapy significantly promotes cell death of HR-HPV infected cells

Summary: This study compares the efficacy of ALA-450 nm PDT and ALA-635 nm PDT and finds that ALA-450 nm PDT is more effective in inhibiting cell proliferation and promoting cell apoptosis than ALA-635 nm PDT, possibly through increasing intracellular ROS generation and caspase-dependent apoptosis pathway. In addition, ALA-450 nm PDT significantly inhibits tumor growth and activates cell apoptosis. Therefore, ALA-450 nm PDT may be a promising therapeutic strategy for eradicating HR-HPV infected cells and HR-HPV related diseases.
