Summary: The study evaluated the clinical value, biological function, and underlying mechanisms of the Cell division cycle associated 5 (CDCA5) in epithelial ovarian cancer (EOC). CDCA5 was found to be upregulated in EOC and correlated with adverse clinicopathological characteristics and poor prognosis. CDCA5 promoted proliferation, invasion, and metastasis while inhibiting apoptosis in EOC cells through the activation of the cell cycle pathway and inhibition of the P53 pathway. Knockdown of CDCA5 expression inhibited malignant activities of EOC cells and suppressed tumor growth in vivo. Mechanistically, the transcription factor KLF5 promoted CDCA5 expression by binding to the CDCA5 promoter. KLF5 overexpression rescued the negative regulation of CDCA5 expression on EOC cell proliferation.

Summary: This study identifies a protein called Gibbin as a key regulator of early epithelial morphogenesis. Gibbin interacts with multiple sequence-specific transcription factors and methyl-CpG-binding proteins to regulate the expression of mesoderm genes. The loss of Gibbin leads to increased DNA methylation at GATA3-dependent mesodermal genes, resulting in a loss of signaling between dermal and epidermal cell types.

Summary: The study on APC gene using a three-dimensional organoid culture of mouse colon epithelia revealed its role in cell proliferation and differentiation regulation, as well as identified novel characteristics related to tumor development and ectopic differentiation. The findings highlight the importance of investigating molecular mechanisms and biological significance of ectopic differentiation in cancer development using colon organoids.

Summary: The study examined the patterns of cell differentiation and proliferation in the liver of alpacas at different stages of development, finding that hepatocytic differentiation began early in development and intensified towards the end of gestation, while cholangiocytic differentiation occurred later. Cell proliferation was observed throughout gestation, indicating similarities in hepatic organogenesis with other species.

Summary: This study investigated the roles of interleukin-1 beta (IL-1 beta) and the p38 mitogen-activated protein kinases (MAPK) signaling pathway in meibomian gland dysfunction (MGD) in rats. It was found that IL-1 beta levels were significantly higher in the terminal ducts of MGD rats compared to young rats. IL-1 beta inhibited cell proliferation, lipid accumulation, and peroxisome proliferator activator receptor gamma (PPAR gamma) expression, and promoted apoptosis by activating the p38 MAPK signaling pathway. SB203580, a specific inhibitor of p38 MAPK, effectively attenuated the effects of IL-1 beta on differentiation, keratinization, and MMP9 expression.

Summary: This study examined the effect of periprostatic adipose tissue supernatant from obese mice on prostate reactivity in vitro and on the viability of human prostatic epithelial cell lines. The results showed that NO and adenosine were involved in the anticontractile and pro-proliferative activities of periprostatic adipose tissue supernatant from obese mice. Further studies are needed to determine whether the blockade of NO and/or adenosine derived from periprostatic adipose tissue can improve prostate function.

Summary: Osteoblasts are crucial in bone modeling and remodeling, with their differentiation being regulated by various signaling pathways. Proper bone homeostasis depends on the differentiation and function of osteoblast lineage cells. Ca2+ signaling controls many processes in osteoblast lineage cells, and K+ channel activation indirectly influences Ca2+ signaling.

Summary: This study reveals that adult stem cell identity and homeostasis are regulated by lineage-restricted transcriptional regulatory networks, with an emphasis on the critical roles of RUNX1 and SMAD3 in maintaining corneal epithelial identity and homeostasis. Furthermore, the cooperation of RUNX1, PAX6, and SMAD3 establishes a core transcription regulatory circuitry for governing lineage-specific cis-regulatory atlas in limbal stem/progenitor cells.

Summary: In renal cell carcinoma (RCC), the interaction between long non-coding RNA (lncRNA) HOTAIR and androgen receptor (AR) forms a feedback loop that promotes tumor angiogenesis, with HOTAIR enhancing GLI2 expression by cooperatively binding to its promoter with AR. These findings suggest that HOTAIR and GLI2 could serve as novel therapeutic targets against RCC.

Summary: Molecular signatures predictive of recurrence-free survival and castration resistance in prostate cancer are crucial for treatment decisions, but current signatures have limited robustness. By analyzing 287 differentially expressed genes, a 6-gene signature called CRPC-derived prognosis signature (CRPCPS) was identified to predict recurrence-free survival and distinguish between castration-resistant and hormone-sensitive prostate cancer. The robustness of this signature was demonstrated in multiple cohorts, showing its potential clinical utility.

Summary: Prostate cancer is a major cause of cancer-related deaths among men, requiring novel treatments. Deslanoside has shown anticancer activity in prostate cancer cells by modulating gene expression, inducing cell cycle arrest at G2/M, and promoting apoptosis. The altered expression of specific genes by deslanoside also correlates with survival outcomes in prostate cancer patients, suggesting potential for its repurposing as an anticancer drug.

Summary: This study compares the efficacy of ALA-450 nm PDT and ALA-635 nm PDT and finds that ALA-450 nm PDT is more effective in inhibiting cell proliferation and promoting cell apoptosis than ALA-635 nm PDT, possibly through increasing intracellular ROS generation and caspase-dependent apoptosis pathway. In addition, ALA-450 nm PDT significantly inhibits tumor growth and activates cell apoptosis. Therefore, ALA-450 nm PDT may be a promising therapeutic strategy for eradicating HR-HPV infected cells and HR-HPV related diseases.
