Summary: A forecasting model combining hidden Markov model and lasso-penalized logistic regression accurately predicts the risk and time course of immune effector cell-associated neurotoxicity syndrome (ICANS) following chimeric antigen receptor (CAR) T-cell therapy. Clinical predictors such as maximum daily temperature, C reactive protein, IL-6, and procalcitonin were used to develop the model. The model showed high accuracy in predicting the occurrence of ICANS and severe ICANS up to 5 days in advance, as well as the entire future risk trajectory from day 5.

Summary: Adoptive therapy using genetically engineered T cells shows promise in treating infectious diseases in immunocompromised individuals. In this study, chimeric engulfment receptor (CER) T cells designed to recognize phosphatidylserine (PS)-expressing cells successfully eliminated SIV-infected cells. The introduction of innate immune functions into T cells enhances the elimination of SIV-infected cells, and further in vivo studies are needed to evaluate their effectiveness.

Summary: IL13R alpha 2 is overexpressed in various cancers, while its binding partner IL13R alpha 1 is widely expressed in healthy tissue. This study generated two variants with improved selectivity for IL13R alpha 2 response using structure-guided protein engineering, and demonstrated their potential application in treating systemic malignancies expressing IL13R alpha 2.

Summary: The study found that CD160 was the most upregulated co-inhibitory receptor in patients with CLL, and its expression was associated with an exhausted T cell phenotype. EVs were identified as a source of CD160 in the plasma of CLL patients. Additionally, a dominantly proinflammatory cytokine profile, especially elevated interleukin-16, was observed in CLL patients' plasma.

Summary: Cancer immunotherapy is more effective against hematological malignancies than solid tumors due to metabolic challenges in the tumor microenvironment. Mitochondria play a key role in CD8(+) T cell activation and effector function. Enhanced mitochondrial function is associated with improved T cell migration and effector function, highlighting the importance of mitochondrial metabolism in T cell responses.

Summary: Elevated levels of circulating immune complexes are associated with autoimmunity and worse prognoses in cancer. IgG-ICs inhibit the proliferation and differentiation of a subset of naive T cells while stimulating the division of another naive-like T cell subset. Phenotypic analysis revealed immature features in the inhibited T cell subset and transcriptional features indicative of a more differentiated phenotype in the stimulated T cell subset.

Summary: The most important feature of adaptive immunity is its specificity, which allows for precise recognition of self, altered-self, and non-self. This specificity is based on millions of years of evolution that resulted in the diversification mechanisms of antigen-recognizing receptors and the emergence of the antigen presentation system. AIRE, a noncanonical transcription factor, plays a crucial role in this system by controlling differentiation and expression of peripheral tissue-specific antigens, as well as increasing the diversity and transfer of presented antigens. This review highlights the importance of the antigen presentation system and discusses the role of AIRE.

Summary: This study provides new information on the efficacy of PD-L1-targeted CAR against PD-L1(low) targets. The results show that PD-L1-CAR cells have strong reactivity and cytotoxicity against both PD-L1(high) and PD-L1(low) target cells. Additionally, PD-L1-CAR cells also exhibit potent cytotoxic effects against non-malignant cells.
