The conceptual significance of understanding functional brain alterations and cognitive deficits associated with Alzheimer's disease (AD) process has been widely established. However, the whole-brain functional networks of AD and its prodromal stage, mild cognitive impairment (MCI), are not well clarified yet. In this study, we compared the characteristics of the whole-brain functional networks among cognitively normal (CN), MCI, and AD individuals by applying graph theoretical analyses to [F-18] fluorodeoxyglucose positron emission tomography (FDG-PET) data. Ninety-four CN elderly, 183 with MCI, and 216 with AD underwent clinical evaluation and FDG-PET scan. The overall small-world property as seen in the CN whole-brain network was preserved in MCI and AD. In contrast, individual parameters of the network were altered with the following patterns of changes: local clustering of networks was lower in both MCI and AD compared to CN, while path length was not different among the three groups. Then, MCI had a lower level of local clustering than AD. Subgroup analyses for AD also revealed that very mild AD had lower local clustering and shorter path length compared to mild AD. Regarding the local properties of the whole-brain networks, MCI and AD had significantly decreased normalized betweenness centrality in several hubs regionally associated with the default mode network compared to CN. Our results suggest that the functional integration in whole-brain network progressively declines due to the AD process. On the other hand, functional relatedness between neighboring brain regions may not gradually decrease, but be the most severely altered in MCI stage and gradually re-increase in clinical AD stages.

Transfer-Tattoo-Like Cell-Sheet Delivery Induced by Interfacial Cell Migration

Ju An Park, Yejin Youm, Hwa-Rim Lee, Yongwoo Lee, Sarah L. Barron, Taejeong Kwak, Gyu Tae Park, Young-Cheol Song, Roisin M. Owens, Jae Ho Kim, Sungjune Jung

Summary: A cell sheet transfer method from a culture surface to a target tissue is introduced in this study. UV-treated parylene, a commercially available flexible material, is used as the culture surface to provide adequate cell adhesiveness for both cell attachment during culture and efficient transfer. Various cell types are successfully transferred using this versatile cell-transfer process. The mechanism of interfacial cell migration between surfaces with different adhesion preferences is observed, and the re-establishment and reversal of cell polarity after transfer are identified. The unique capabilities of 3D multilayer stacking, freeform design, and curved surface application are demonstrated. The therapeutic potential of the cell-sheet delivery system is demonstrated through experiments on cutaneous wound healing and skin-tissue regeneration in mice models.

Functional expression of oxytocin receptors in pulp-dentin complex

Ju Won Jung, So Young Park, Eun Jin Seo, Il Ho Jang, Yeji Park, Dasun Lee, Dohyun Kim, Jin Man Kim

Summary: Dental pulp-derived stromal cells (DPSCs) are important for maintaining the tissue integrity of the pulp-dentin complex. In this study, we demonstrate the biological functions and significance of the oxytocin receptor (OXTR) in DPSCs. Blocking OXTR activity promotes DPSC differentiation and is involved in extracellular matrix (ECM) remodeling. The genetic changes mediated by OXTR are regulated by the transcriptional activity of Yes-associated protein (YAP). These findings provide new insights into the regenerative process of the pulp-dentin complex.

Summary: The study aimed to evaluate the diagnostic and prognostic value of non-hyperemic Pd/Pa and its additional value when combined with hyperemic pressure ratio (FFR) to guide revascularization. A total of 2141 patients with physiological data in 2726 vessels were included in a retrospective registry. Abnormal pressure ratios in both non-hyperemic and hyperemic conditions were found to have important prognostic value. The combined use of FFR and non-hyperemic Pd/Pa efficiently identified vessels with concordant normal pressure ratios and excellent long-term clinical outcomes.

Summary: This study evaluated the association between cardiac diastolic dysfunction and coronary microvascular dysfunction (CMD) in patients without significant left ventricular systolic dysfunction and epicardial coronary stenosis. The study found that CMD was independently associated with cardiac diastolic dysfunction and both were associated with an increased risk of cardiovascular death or admission for heart failure.

TRRAP Enhances Cancer Stem Cell Characteristics by Regulating NANOG Protein Stability in Colon Cancer Cells

Kyung-Taek Kang, Min-Joo Shin, Hye-Ji Moon, Kyung-Un Choi, Dong-Soo Suh, Jae-Ho Kim

Summary: This study found that TRRAP overexpression increases NANOG protein stability and enhances the tumorigenic potential of colon cancer stem cells. TRRAP inhibits the ubiquitination degradation of NANOG by binding to it. TRRAP knockdown decreases the expression of CD44, a cancer stem cell marker, and increases the expression of P53, a tumor suppressor gene, in colon cancer cells.

Physicochemical Properties and Biocompatibility of Various Bioceramic Root Canal Sealers: In Vitro Study

Sang Won Kwak, Jina Koo, Minju Song, Il Ho Jang, Gianluca Gambarini, Hyeon-Cheol Kim

Summary: The aim of this study was to compare the physicochemical properties and biocompatibility between various calcium silicate-based bioceramic sealers (CSBSs) and the epoxy resin-based sealer AH Plus (AHP). The physical properties of CSBSs, including flow, setting time, radiopacity, dimensional stability, and pH, were evaluated according to the ISO 6876 standards. The biocompatibility of CSBSs was found to be higher than that of AHP, as assessed through cytotoxicity and cell attachment assays.

Summary: This study aimed to determine if the addition of PET/MRI and CA 19-9 to CECT can improve the accuracy of predicting resectability and prognosis in PDAC patients. The results showed that the combination of CECT, PET, and CA 19-9 increased the AUC and sensitivity for determining resectability without compromising specificity. Additionally, FDG avidity on post-NAT PET and pathologically confirmed vascular invasion predicted recurrence-free survival in PDAC patients.

Sonazoid-enhanced ultrasonography for noninvasive imaging diagnosis of hepatocellular carcinoma: special emphasis on the 2022 KLCA-NCC guideline

Hyo-Jin Kang, Jeong Min Lee, Se Woo Kim

Summary: Contrast-enhanced ultrasonography (CEUS) is a noninvasive imaging modality used for diagnosing hepatocellular carcinoma (HCC) without the need for pathological confirmation. Different contrast agents, such as SonoVue and Sonazoid, are available for CEUS imaging. Major guidelines recognize CEUS as a reliable method for HCC diagnosis, but the choice of contrast agent varies. This review provides a comparative overview of the pharmacokinetic features, examination protocols, diagnostic criteria, and potential applications of these contrast agents in the HCC diagnostic algorithm.

Summary: Coronary stent underexpansion is associated with restenosis and stent thrombosis. High wall shear stress (WSS) has been linked to prothrombotic pathways activation, matrix metalloproteinases upregulation, and future myocardial infarction in atherosclerosis studies. This study aimed to investigate the relationship between stent underexpansion and WSS. The results showed that frames with stent underexpansion were more likely to exhibit high WSS, while no significant association was found between underexpansion and low WSS or between underexpansion and stent platform.
