Summary: Nucleic acids have great potential for gene therapy, with plasmid DNA as the first therapeutic nucleic acid and mRNA as a safer and more affordable option. This study investigated the uptake mechanisms and efficiencies of genetic material by cells, focusing on different nucleic acids, delivery vectors, and human primary cells. The study found that caveolae-mediated uptake was the main route for gene delivery, pDNA had higher expression levels in fast-dividing fibroblasts, cmRNA was responsible for high protein production in slow-dividing osteoblasts, and in mesenchymal stem cells, the combination of vector and nucleic acid was more relevant. Protein expression was higher when cells were seeded on 3D scaffolds.

Summary: Abdominal sepsis leads to the migration of microorganisms from the gut to the peritoneum and bloodstream, but there are limited methods and biomarkers to study and monitor this process. In this study, cecal ligation and puncture (CLP) were used to induce abdominal sepsis in mice, and NGS was employed to analyze the microbial species composition in fecal, peritoneal lavage, and blood samples. The results showed rapid changes in gut microbial communities after CLP, with pathogenic species detected in the peritoneum and blood as early as 24 hours post-CLP. NGS of cfDNA from blood samples as small as 30 microliters was able to accurately identify pathogenic species in a time-dependent manner.

Summary: Recently, a surgical suction filter device was introduced to generate a suction filter-derived bone grafting substitute (SF-BGS). This study aimed to investigate the quality of the SF-BGS by determining the expression of miRNAs and osteogenic mRNAs. The results showed distinct differences in miRNA expression and overall upregulation of osteogenic marker genes in SF-BGS.

Summary: In this study, the miRNA expression profile in enthesis tissue following injury was investigated. Target prediction and mRNA expression analysis suggest that certain miRNAs may regulate the expression of mRNA targets relevant for enthesis healing. Additionally, the protein expression levels of collagens I, II, III, and X were found to change after injury.

Summary: This study investigates the effects of Early Total Care (ETC) and Damage Control Orthopedics (DCO) on microRNA (miRNA) expression in extracellular vesicles (EVs) and identifies their mRNA targets. The results show that the miRNA expression patterns in the ETC group are more pro-inflammatory, indicating potentially decreased tissue regeneration capabilities. In silico target prediction reveals mRNA targets related to inflammation, fibrosis, and Wnt signaling. Validation of these targets in lung tissue shows treatment-specific deregulations that match the expression of their upstream miRNAs, while no significant deregulations were observed in the liver.

Summary: Low-dose irradiation is an effective treatment option for persistent lymphatic fistulas after inguinal vascular surgery, preventing infection, sepsis, bleeding, and possible leg amputation. The recommended dose ranges from 0.3-0.5 to 1-12 Gy. In this study, every patient experienced resolution of the lymphatic fistula without major complications.

Summary: Various cell surface receptors, including cadherin-11 and platelet-derived growth factor receptors (PDGFRs), are involved in the differentiation and self-renewal of human mesenchymal stem cells (hMSCs). This study used a computational model to investigate the interactions between cadherin-11 and PDGFRs, and revealed the existence of crosstalk at both receptor and downstream signaling levels. The findings contribute to a better understanding of the effect of cell surface receptors on hMSCs proliferation.
