Antiplatelet Agents Can Promote Two-Peaked Thrombin Generation in Platelet Rich Plasma: Mechanism and Possible Applications

Background: Thrombin generation assay is a convenient and widely used method for analysis of the blood coagulation system status. Thrombin generation curve (TGC) is usually bell-shaped with a single peak, but there are exceptions. In particular, TGC in platelet-rich plasma (PRP) can sometimes have two peaks. Objective: We sought to understand the mechanism underlying the occurrence of two peaks in the PRP thrombin generation curve. Methods: Tissue factor-induced thrombin generation in PRP and platelet-poor plasma (PPP) was monitored using continuous measurement of the hydrolysis rate of the thrombin-specific fluorogenic substrate Z-Gly-Gly-Arg-AMC. Expression of phosphatidylserine (PS) and CD62P on the surface of activated platelets was measured by flow cytometry using corresponding fluorescently labeled markers. Results: The addition of the P2Y12 receptor antagonist MeS-AMP (160 mu M), 83 nM prostaglandin E-1 (PGE(1)), or 1.6% DMSO to PRP caused the appearance of two peaks in the TGC. The PS exposure after thrombin activation on washed platelets in a suspension supplemented with DMSO, PGE(1) or MeS-AMP was delayed, which could indicate mechanism of the second peak formation. Supplementation of PRP with 1.6% DMSO plus 830 nM PGE(1) mediated the disappearance of the second peak and decreased the amplitude of the first peak. Increasing the platelet concentration in the PRP promoted the consolidation of the two peaks into one. Conclusions: Procoagulant tenase and prothrombinase complexes in PRP assemble on phospholipid surfaces containing PS of two types - plasma lipoproteins and the surface of activated platelets. Thrombin generation in the PRP can be two-peaked. The second peak appears in the presence of platelet antagonists as a result of delayed PS expression on platelets, which leads to delayed assembly of the membrane-dependent procoagulant complexes and a second wave of thrombin generation.

The Role of Immobilized Phospholipids in the Initiation of Blood Coagulation under Flow Conditions

Summary: This study found that plasma coagulation under arterial shear rates can be initiated by the presence of an immobilized phospholipid fraction in the damaged vascular wall. The binding of coagulation factors to these phospholipids protects the initial stages of plasma coagulation from flow and leads to rapid thrombin production even under arterial blood shear rates.

Ex vivo observation of granulocyte activity during thrombus formation

Summary: In this study, an ex vivo experimental model for observation of granulocyte activity in thrombus formation was developed. Results showed that granulocytes from patients with WAS had reduced motility compared to healthy donors. The study suggests that WASP deficiency disrupts granulocyte behavior in thromboinflammatory conditions.

Ammonium removal by erythrocyte-bioreactors based on glutamate dehydrogenase from Proteus sp. jointly with porcine heart alanine aminotransferase

Summary: Novel erythrocyte-bioreactors containing ALT and non-aggregating GDH were successfully produced using the flow dialysis method and the new bacterial GDH enzyme from Proteus sp. The ammonium removal rate increased linearly with an increase in encapsulated GDH activity and alanine production correlated with ammonium consumption, indicating the joint functioning of both enzymes. This suggests that the new bioreactors are promising for clinical use as they have more efficient GDH encapsulation and properties comparable to previously obtained bioreactors.

Thrombus Inhibition and Neuroprotection for Ischemic Stroke Treatment through Platelet Regulation and ROS Scavenging

Summary: In this study, nanodelivery systems based on MnO2 loaded with Ginkgolide B (GB) were designed to restore the intracerebral microenvironment in ischemic stroke. The study found that GB can protect brain tissue from oxidative damage by activating the Nrf2 signaling pathway. Meanwhile, MnO2 nanoparticles can penetrate the blood-brain barrier and enhance the delivery of therapeutic agents within brain tissue. This study provides a new therapeutic route for regulating the microenvironment of ischemic stroke.

Summary: A convenient strategy for the synthesis of pyrroles lineary linked to 4H-pyrrolo[3,2,1-ij]quinolin-2(1H)-ones is presented. The cyclization of (E)-1-(2-oxo-2-phenylethylidene)-4H-pyrrolo[3,2,1-ij]quinolin-2-ones with linear and cyclic enaminones proceeds both in the presence of p-toluenesulfonic acid and triethylamine. The obtained compounds are characterised by atropoisomerism and keto-enol tautomerism. The synthesized 1-(2-phenyl-1H-pyrrol-3-y1)-4H-pyrrolo[3,2,1-ij]quinolin-2(1H)-ones were studied for anticoagulant activity.

In vitro testing showed that these derivatives have the potential anticoagulant activity by inhibiting both coagulation factors Xa and XIa.

These processes occur on specialized scaffolds, such as phospholipid membranes, fibrin and extracellular matrix protein complexes, and polymers containing phosphate groups. The physiological significance and physical meaning of these scaffolds vary, and their roles in hemostasis and thrombosis are still being studied.

Procoagulant Activity in Amniotic Fluid Is Associated with Fetal-Derived Extracellular Vesicles
