Summary: Calcific aortic valve stenosis (CAVS), the third most prevalent cardiovascular disorder, imposes a significant burden on patients but lacks pharmaceutical treatment options. Aortic valve replacement is the only option, but its effectiveness is uncertain and complications are common. Capsaicin, known for its anti-inflammatory and antioxidant properties, has been found to inhibit arterial calcification. In this study, capsaicin was shown to reduce calcium deposition in calcified aortic valve interstitial cells (VICs) and downregulate calcification markers, suggesting its potential as a treatment for CAVS. The mechanism of action involves inhibition of the NF kappa B/AKT/ERK1/2 signaling pathway, oxidative stress, and AGE-RAGE signaling pathways.

Summary: This study found that LIMD2 is up-regulated in ovarian cancer tissues and cells, and its knockdown can inhibit the proliferation, migration, invasion, and autophagy of ovarian cancer cells, while promoting apoptosis. The inhibition of LIMD2 is achieved through the ERK1/2 signaling pathway. Therefore, LIMD2-siRNA may be an effective molecule to prevent ovarian cancer progression.

MiR-100-5p in NONFH exosomes targets BMPR2 and suppresses the BMPR2/SMAD1/5/9 signaling pathway, leading to NONFH-like damage. Silencing miR-100-5p expression rescues the reduction in osteogenesis and angiogenesis by activating the BMPR2/SMAD1/5/9 signaling pathway.

Summary: The present study investigated the effect of T-2 toxin on osteoblast differentiation and mineralization by treating MC3T3-E1 cells with different concentrations of T-2 toxin. The results showed that T-2 toxin at 4 and 8 nM significantly inhibited cell viability and impaired osteoblastic differentiation and mineralization. The activation of the Wnt signaling pathway mitigated the T-2 toxin-induced damage, while the inhibition of autophagy exacerbated it. Our findings also revealed a positive feedback loop between the Wnt signaling pathway and autophagy.
