Summary: This study investigated whether the analysis of EV-miRNA is affected by the presence of platelets and ery-ghosts in human plasma. The results showed that filtration of plasma can remove platelets and ery-ghosts, improving the comparability and reproducibility of EV-miRNA studies. The authors recommend measuring and reporting the plasma concentration of platelets for EV-miRNA studies, and filtering plasma before downstream analyses or storage in biobanks.

Summary: T cells play a central role in inflammatory diseases, but we still have limited knowledge about the TCR repertoires in tissues affected by these diseases. By examining autopsies from patients with and without GVHD and using mouse models of GVHD, we found that the composition of TCR repertoires in tissues was mainly determined by anatomical similarity, not by the disease or transplant status. T cells in peripheral blood and spleens only captured a small portion of the TCR repertoires detected in tissues, and motif-based clustering revealed shared repertoire signatures across patients in a tissue-specific manner. The tissue-resident T cells at disease sites were of donor origin. These findings highlight the importance of studying T cells in tissues for tissue-based pathologies and indicate the tissue-specific nature of both endogenous and posttransplant T cell populations.

Summary: In this study, a CD154 upregulation assay was used to quantify Ni2+, Co2+, and Pd2+ specific CD4+ T cells, and the related TCR repertoire was analyzed by high-throughput sequencing. The results revealed an overexpression of the gene segment TRAV9-2 in Ni2+ specific TCRs, while Co2+ and Pd2+ specific TCRs favored the expression of other gene segments. Furthermore, a histidine in the CDR3 of metal-specific TCRs was found to be conserved among cross-reactive clonotypes.

Summary: This study compares different density and velocity gradients, along with commercial kits, for separating EVs from HIV particles and contaminating protein, to evaluate the suitability of current plasma EV analysis techniques in identifying new biomarkers of HIV-1 immune activation. The findings reveal the optimal separation of EVs from viruses and free proteins using iodixanol velocity gradient, and the identification of a new population of large EVs enriched in miR-155 and mitochondrial DNA, paving the way for a deeper understanding of HIV-1 pathogenesis.
