Translation Directed by Hepatitis A Virus IRES in the Absence of Active eIF4F Complex and eIF2

Translation directed by several picornavirus IRES elements can usually take place after cleavage of eIF4G by picornavirus proteases 2A(pro) or L-pro. The hepatitis A virus (HAV) IRES is thought to be an exception to this rule because it requires intact eIF4F complex for translation. In line with previous results we report that poliovirus (PV) 2A(pro) strongly blocks protein synthesis directed by HAV IRES. However, in contrast to previous findings we now demonstrate that eIF4G cleavage by foot-and-mouth disease virus (FMDV) L-pro strongly stimulates HAV IRES-driven translation. Thus, this is the first observation that 2A(pro) and L-pro exhibit opposite effects to what was previously thought to be the case in HAV IRES. This effect has been observed both in hamster BHK and human hepatoma Huh7 cells. In addition, this stimulation of translation is also observed in cell free systems after addition of purified L-pro. Notably, in presence of this FMDV protease, translation directed by HAV IRES takes place when eIF2 alpha has been inactivated by phosphorylation. Our present findings clearly demonstrate that protein synthesis directed by HAV IRES can occur when eIF4G has been cleaved and after inactivation of eIF2. Therefore, translation directed by HAV IRES without intact eIF4G and active eIF2 is similar to that observed with other picornavirus IRESs.

Summary: The aim of this study was to identify serum miRNAs that can phenotype asthma patients. Next-generation sequencing and RT-qPCR validation revealed that hsa-miR-26a-1-3p and hsa-miR-376a-3p were associated with eosinophilic and noneosinophilic asthma. Furthermore, the expression of these miRNAs was correlated with the pathogenesis of asthma.

Viruses, friends, and foes: The case of Torque Teno Virus and the net state of immunosuppression

Summary: New reliable biomarkers are needed to predict immune-related complications after organ and stem cell transplantation. Torque Teno Virus (TTV) shows promise as a functional surrogate for monitoring immunosuppression, with high prevalence rates and stable viral loads. Studies suggest that increasing TTV DNA levels are associated with infectious complications, while decreasing levels are associated with acute rejection. However, the interpretation in stem cell transplant recipients is complicated by various factors. Standardization of PCR methods and ongoing trials evaluating a TTV load-guided strategy are expected in the future.

Dynamics of Human Anelloviruses in Plasma and Clinical Outcomes Following Kidney Transplantation

Summary: This study investigates the genetic diversity and clinical correlates of the Anelloviridae family after kidney transplantation. The dominant genus was Alphatorquevirus, followed by Betatorquevirus and Gammatorquevirus. Changes in the predominant genus were observed in some patients. Overall, the levels of Anelloviridae DNA increased after transplantation and were associated with the use of T-cell depleting agents. However, there were no associations between the dominant genus or total Anelloviridae DNA levels and clinical outcomes.

Summary: Clonal evolution in acute myeloid leukemia (AML) can be reconstructed using integrated multidimensional flow cytometry (MFC) immunophenotyping and fluorescence-activated cell sorting (FACS) with next-generation sequencing (NGS). Mutational analysis of dysplastic cells and leukemic blasts at the onset of AML identified stable, branching, and clonal evolution patterns. Techniques other than single-cell multiomics can be used to detect measurable residual disease (MRD) and genetic traits associated with treatment resistance in patients achieving complete response (CR). Whole-exome sequencing revealed different clonal involvement in dysplastic myeloerythropoiesis, leukemic transformation, and chemoresistance.

Summary: Applications of Structural Health Monitoring (SHM) combined with Machine Learning (ML) techniques enhance real-time performance tracking and increase structural integrity awareness of civil, aerospace and automotive infrastructures. In this paper, a novel ML algorithm is developed to predict fatigue damage in composite materials using the signals of Lamb Waves and other meaningful features. The proposed algorithm achieves good accuracy and computation times comparable to other ML algorithms.

Summary: This study compares two methods (intracellular cytokine staining and interferon-gamma release assay) in predicting immune protection among kidney transplant recipients. Summary: The goal of this research is to introduce a novel tunable 3D metamaterial that can reproduce a wide spectrum of 3D auxetic and non-auxetic Poisson's ratios and Young's moduli. By changing the connectivity and position of the cell's internal nodes, this metamaterial can accommodate different spatial curvatures and boundaries. This wide range of variation is achieved through simple spatial triangularization and allows for better adaptability to component design boundaries and shapes.

The Updated Properties Model (UPM): A topology optimization algorithm for the creation of macro-micro optimized structures with variable stiffness

Summary: 3D printing is changing the design and manufacturing of high value industrial components, pursuing a macro-micro structural optimization of architected metamaterials to customize mechanical properties. Topology optimization techniques facilitate the design of microstructures and overall component topology. A novel topology optimization algorithm is proposed, aiming to homogenize the strain level by minimizing its standard deviation, without imposing constraints and reducing external parameters. The proposed technique, working directly with moduli, is suitable for material-component design using functionally graded metamaterials and can be extended to non-linear isotropic materials.

Summary: Previous statin therapy had no apparent protective effect on the outcome of post-transplant bloodstream infection among kidney transplant recipients. The time to clinical cure and secondary outcomes such as fever resolution, ICU admission or renal replacement therapy requirement, and 30-day all-cause mortality did not differ significantly between statin and non-statin groups.

Summary: The study found that SARS-CoV-2 encodes eleven accessory proteins in its genome, and their roles during infection are not fully understood. Transcriptomics analysis revealed that WNT5A and IL11 were significantly up-regulated in A549 cells expressing certain accessory proteins from SARS-CoV-2. Bioinformatics analysis and functional assays confirmed the involvement of WNT5A and IL11 in pulmonary fibrosis, and altered profibrotic gene expression was observed in lung cell lines infected with SARS-CoV-2 and COVID-19 patients' lung biopsies. These findings suggest that targeting these accessory proteins could be a potential therapeutic approach against COVID-19.

The analysis of 465 KT recipients revealed an association between CMV viremia and the incidence of post-transplant atherosclerotic (PAEs) and thrombotic events (PTEs), but this association was not significant after adjusting for various factors.
