Synchrotron Ultraviolet Microspectroscopy on Rat Cortical Bone: Involvement of Tyrosine and Tryptophan in the Osteocyte and Its Environment

Alcohol induced osteoporosis is characterized by a bone mass decrease and microarchitecture alterations. Having observed an excess in osteocyte apoptosis, we aimed to assess the bone tissue biochemistry, particularly in the osteocyte and its environment. For this purpose, we used a model of alcohol induced osteoporosis in rats. Bone sections of cortical bone were investigated using synchrotron UV-microspectrofluorescence at subcellular resolution. We show that bone present three fluorescence peaks at 305, 333 and 385 nm, respectively corresponding to tyrosine, tryptophan and collagen. We have determined that tyrosine/collagen and tryptophan/collagen ratios were higher in the strong alcohol consumption group. Tryptophan is related to the serotonin metabolism involved in bone formation, while tyrosine is involved in the activity of tyrosine kinases and phosphatases in osteocytes. Our experiment represents the first combined synchrotron UV microspectroscopy analysis of bone tissue with a quantitative biochemical characterization in the osteocyte and surrounding matrix performed separately.

Investigations by AFM of Ageing Mechanisms in PLA-Flax Fibre Composites during Garden Composting

Summary: PLA-flax non-woven composites show preferential degradation of flax fibres from the edge to the core during garden composting, while erosion of cell walls occurs within the fibres themselves from the central lumen to the periphery primary wall. The ageing process results in a decrease in indentation modulus for fibres in the preserved core of the composite and a change in PLA morphology towards the edge of the sample.

Fluorescence microscopy and photodielectric characterization studies of the composite films of polyvinyl alcohol and tryptophan functionalized silver nanoparticles

Summary: In this study, fluorescent nanocomposite films were prepared by solution mixing of polyvinyl alcohol (PVA) and tryptophan functionalized silver (AgTrp) nanoparticles. The distribution of AgTrp nanoparticles within the polymer matrix was tracked using synchrotron excitation deep-ultraviolet fluorescence imaging. The presence of AgTrp nanoparticles resulted in the pronounced growth of dendrite-like structures, which was confirmed by various imaging techniques. Both tryptophan and AgTrp nanoparticles affected the thermal properties of the matrix, reducing crystallinity and increasing the glass transition temperature of PVA. Furthermore, the films showed a significant increase in conductance under illumination with 250 nm light, with the relative changes reaching almost 200% compared to the dark condition.
