Summary: Through a mouse forward genetic screening, we identified that one of the key Wnt/β-catenin effectors TCF7L2/TCF4 controls vocalization production and complexity. The early developmental expression of TCF7L2 in PAG excitatory neurons is necessary for this trait, and TCF7L2-mediated vocal control is dependent on its DNA binding ability. Patient mutations associated with developmental disorders disrupt the transcriptional repression effect of TCF7L2, leading to severe impairments in vocalization.

Summary: Pancreatic adenocarcinoma (PAAD) is a highly aggressive cancer. RNA-binding protein STAU2 plays a critical role in the occurrence and development of PAAD, and it can be a potential biomarker and therapeutic target for PAAD. High expression of STAU2 is associated with poor clinical outcomes and immune evasion in PAAD. Additionally, STAU2 is related to chemotherapy drug sensitivity, suggesting it could be used in combination therapy. This study provides valuable insights into the role of STAU2 in PAAD aggression and resistance, and it sheds light on its potential for cancer therapy and drug development.

Summary: Hyaluronic acid, a component of the central nervous system (CNS), has been found in lesions of multiple sclerosis (MS) patients and mouse models of autoimmune encephalomyelitis (EAE). This study demonstrates that hyaluronic acid and its receptor CD44 play a role in inhibiting the differentiation of oligodendroglial cells, and this effect requires the involvement of transmembrane protein 2 (TMEM2), a cell surface hyaluronidase. These findings provide insights into the physiological and pathological effects of hyaluronic acid on oligodendroglial cells.

LDB1 is a key factor in the transformation of thymocytes into T-ALL induced by LMO2 and enhances the stability of cancer-related proteins in leukemia.

Summary: In this study, we conducted a comprehensive screening of genes involved in primitive streak (PrS) formation and identified 812 genes that may be linked to this process. We discovered that elevated intracellular ceramide negatively regulates gene expression essential for PrS formation and instead induces neurogenesis. Additionally, sphingosine-1-phosphate positively regulates neural maturation. These findings highlight the role of ceramide in regulating both PrS formation and neural differentiation.
