Summary: In this study, Vclb was found to inhibit the NF-KB signaling pathway, and its inhibitory effect was enhanced by LPS induction. Furthermore, Vclb inhibits NF-KB activation by targeting MyD88, thereby suppressing the production of inflammatory cytokines. Mechanistically, Vclb inhibits the NF-KB signaling pathway by targeting MyD88 ubiquitin-proteasome pathway.

Summary: The study reveals that ADAR-mediated RNA editing may be an important mechanism underlying genetic variants associated with common inflammatory diseases, with disease risk variants associated with reduced editing of cellular double-stranded RNA (dsRNA) and induction of interferon responses and inflammation. This suggests that cellular dsRNA editing and sensing could be an underappreciated mechanism of common inflammatory diseases.

Summary: Diabetic kidney disease (DKD) is a common complication of diabetes, and the understanding of its molecular mechanisms is still incomplete. A recent study found that the expression of deubiquitinating enzyme USP25 was significantly upregulated in the kidneys of diabetic mice, and its absence worsened diabetes-induced renal dysfunction and fibrosis.

Summary: The host engulfment protein ELMO1 interacts with WxxxE effectors like SifA, influencing intestinal inflammation and bacterial pathogenicity. The ELMO1-WxxxE effector interaction plays a crucial role in regulating bacterial colonization, dissemination, and induction of inflammatory cytokines. It has been found that the interaction between ELMO1 and WxxxE motif-containing effectors, such as SifA, not only affects the pathogenicity of enteric pathogens but also plays a role in modulating inflammatory responses and cellular activities.

Summary: CARD8, a pattern-recognition receptor, is involved in the activation of the inflammasome. The degradation of CARD8 by the 20S proteasome plays a crucial role in the control of inflammasome activation.

Summary: This study found that the Avibirnavirus VP3 protein is ubiquitinated during infection, enhancing virus replication. VP3 inhibits the immune response and evades the host immune system by autophagy. These findings provide insights into potential drug targets for controlling virus infection.

Summary: Mitochondria play versatile roles in cellular processes and have emerged as critical components in innate immunity. This review provides a comprehensive exploration of the multifaceted mechanisms underlying the interactions between mitochondria and innate immune responses. It discusses the roles of healthy mitochondria as platforms for signalosome assembly, the release of mitochondrial components as signaling messengers, and the regulation of signaling via mitophagy. Additionally, it explores the impacts of mitochondrial proteins and metabolites on modulating innate immune responses, immune cell polarization, and their implications in infectious and inflammatory diseases.

Summary: This study reveals the functional compensation and complementation of IRAK1 in leukemic cells upon inhibition of IRAK4. Cotargeting IRAK1 and IRAK4 is necessary to suppress leukemic stem/progenitor cell function and induce differentiation. IRAK1 and IRAK4 maintain the undifferentiated state of MDS/AML LSPCs through noncanonical MyD88-independent pathways.

Summary: Macrophages play a critical role in immune homeostasis and host defense, but their uncontrolled activation can lead to tissue injury and autoimmunity. This study investigates the molecular mechanisms of macrophage activation and identifies lysine-specific demethylase 5B (KDM5B) as a key player in the activation of NF-kappa B signaling cascade and pro-inflammatory cytokine production. KDM5B deficiency or inhibition protects against immunologic injury in animal models, suggesting it as a potential therapeutic target for autoimmune and inflammatory disorders.
