Validation of Reference Genes for the Relative Quantification of Gene Expression in Human Epicardial Adipose Tissue

Background: Relative quantification is a commonly used method for assessing gene expression, however its accuracy and reliability is dependent upon the choice of an optimal endogenous control gene, and such choice cannot be made a priori. There is limited information available on suitable reference genes to be used for studies involving human epicardial adipose tissue. The objective of the current study was to evaluate and identify optimal reference genes for use in the relative quantification of gene expression in human epicardial fat depots of lean, overweight and obese subjects. Methodology/Principal Findings: Some of the commonly used reference genes including 18S, ACTB, RPL27, HPRT, CYCA, GAPDH, RPLPO, POLR2A and B2M were quantified using real-time PCR analysis. The expression stability of these genes was evaluated using Genorm, Normfinder and Bestkeeper algorithms. In addition, the effect of sample size on the validation process was studied by randomly categorizing subjects in two cohorts of n = 2 and n = 33. Conclusions/Significance: CYCA, GAPDH and RPL27 were identified as the most stable genes common to all three algorithms and both sample sizes. Their use as reference gene pairs might contribute to the enhanced robustness of relative quantification in the studies involving the human epicardial adipose tissue.

Summary: Multi-omic profiling of patients with coronary artery disease reveals links between alterations in the serum metabolome with genetics, diet, and the microbiome. The study found that these metabolomic alterations are associated with metabolic impairment preceding clinically overt coronary artery disease. The results highlight the importance of understanding risk-factor heterogeneity in coronary artery disease through the serum metabolome.

Summary: This study investigates the alterations of the metabolome and microbiome from dysmetabolic conditions to ischemic heart disease (IHD). The findings suggest that major changes in the gut microbiome and metabolome may occur long before the clinical onset of IHD. Specific microbiome and metabolome features associated with IHD could better differentiate individuals with IHD from healthy individuals, indicating their pathophysiological relevance.

Summary: Variants within dominant SARS-CoV-2 T cell epitopes have been identified through global sequence data analysis. These variants, independently arising in multiple lineages, result in loss of T cell recognition of specific epitopes. The findings demonstrate the potential for T cell evasion and emphasize the importance of ongoing surveillance for variants capable of escaping immune responses.

Summary: This study found that complications and worse functional outcomes in patients admitted to hospital with COVID-19 are high, even in young, previously healthy individuals. Acute complications are associated with reduced ability to self-care at discharge, with neurological complications being associated with the worst functional outcomes. COVID-19 complications are likely to cause a substantial strain on health and social care in the coming years.
