MicroRNA-141 Represses HBV Replication by Targeting PPARA

MicroRNAs (miRNAs) are small non-coding RNAs that regulate gene expression primarily at the post-transcriptional level and play critical roles in a variety of physiological and pathological processes. In this report, miR-141 was identified to repress HBV expression by screening a small miRNA expressing library and synthetic miR-141 mimics could also significantly suppress HBV expression and replication in HepG2 cells. Bioinformatic analysis and experiment assays indicate that peroxisome proliferator-activated receptor alpha (PPARA) was the target of hsa-miR-141 during this process. Furthermore, knockdown of PPARA by small interfering RNA (siRNA) inhibited HBV replication similar to levels observed for miR-141. Promoter functional analysis indicated that repression of HBV replication by miR-141 mimics or siRNA was mediated by interfering with the HBV promoter functions, consistent with previous studies demonstrating that PPARA regulated HBV gene expression through interactions with HBV promoter regulatory elements. Our results suggest that miR-141 suppressed HBV replication by reducing HBV promoter activities by down-regulating PPARA. This study provides new insights into the molecular mechanisms associated with HBV-host interactions. Furthermore, this information may facilitate the development of novel anti-HBV therapeutic strategies.

Identification of Montelukast as flavivirus NS2B-NS3 protease inhibitor by inverse virtual screening and experimental validation

Summary: Flavivirus, including Dengue Virus (DENV) and Zika virus (ZIKV), infects and kills millions of people every year, but there is no approved treatment available. In this study, we investigated the inhibitory mechanisms of Montelukast, an existing drug, against the replication of DENV and ZIKV. Through computational methods and in vitro experiments, we found that Montelukast can bind to NS2B-NS3 proteases of DENV and ZIKV and act as a competitive inhibitor. This research provides new insights for the development of potential antiviral drugs.

Summary: In this study, a new strategy of incorporating glycyrrhizic acid (GA) and polyene phosphatidylcholine (PPC) into lipid nanoparticles (GA/PPC-modified LNPs) was developed, which showed enhanced cellular uptake, improved gene-silencing efficiency, reduced cytotoxicity, and increased siRNA stability. GA/PPC-modified LNPs targeting NF-kappa B attenuated acute liver injury and facilitated intracellular delivery of antisense oligonucleotides (ASOs) and mRNA inhibiting viral infection. These findings suggest that GA/PPC-modified LNPs could serve as a promising nucleic acid delivery system.

Durability of neutralization against Omicron subvariants after vaccination and breakthrough infection

Summary: A study found that the durability of neutralizing antibodies against the Omicron subvariants is uncertain 4-6 months after vaccination or infection. Two doses of CoronaVac or a third dose of ZF2001 produce limited neutralization against Omicron subvariants 6 months after vaccination. Hybrid immunity and Delta, BA.1, and BA.2 breakthrough infections induce long-term persistence of the antibody response, with over 70% of sera neutralizing BA.1, BA.2, BA.4/BA.5, and BF.7. However, BQ.1.1 and XBB, as well as BA.2.75, show higher resistance to neutralization. These data suggest the need for additional vaccination in CoronaVac or ZF2001-vaccinated individuals and provide insight into the durability of neutralization against Omicron subvariants.
