PLGA, PLGA-TMC and TMC-TPP Nanoparticles Differentially Modulate the Outcome of Nasal Vaccination by Inducing Tolerance or Enhancing Humoral Immunity

Development of vaccines in autoimmune diseases has received wide attention over the last decade. However, many vaccines showed limited clinical efficacy. To enhance vaccine efficacy in infectious diseases, biocompatible and biodegradable polymeric nanoparticles have gained interest as antigen delivery systems. We investigated in mice whether antigen-encapsulated PLGA (poly-lactic-co-glycolic acid), PLGA-TMC (N-trimethyl chitosan) or TMC-TPP (tri-polyphosphate) nanoparticles can also be used to modulate the immunological outcome after nasal vaccination. These three nanoparticles enhanced the antigen presentation by dendritic cells, as shown by increased in vitro and in vivo CD4(+) T-cell proliferation. However, only nasal PLGA nanoparticles were found to induce an immunoregulatory response as shown by enhanced Foxp3 expression in the nasopharynx associated lymphoid tissue and cervical lymph nodes. Nasal administration of OVA-containing PLGA particle resulted in functional suppression of an OVA-specific Th-1 mediated delayed-type hypersensitivity reaction, while TMC-TPP nanoparticles induced humoral immunity, which coincided with the enhanced generation of OVA-specific B-cells in the cervical lymph nodes. Intranasal treatment with Hsp70-mB29a peptide-loaded PLGA nanoparticles suppressed proteoglycan-induced arthritis, leading to a significant reduction of disease. We have uncovered a role for PLGA nanoparticles to enhance CD4(+) T-cell mediated immunomodulation after nasal application. The exploitation of this differential regulation of nanoparticles to modulate nasal immune responses can lead to innovative vaccine development for prophylactic or therapeutic vaccination in infectious or autoimmune diseases.

Summary: CR-163 and CR-172 exhibit broad-spectrum antimicrobial activity by disrupting bacterial membranes. Additionally, PepBiotics can bind to LPS and LTA, replacing the role of polymyxin B. The study also demonstrates that bacteria killed by PepBiotics are less likely to stimulate immune cells compared to gentamicin and heat-killed bacteria, which still elicit a strong immune response.

Summary: This study investigated the effects of knee joint distraction (KJD) on osteoarthritic joints in dogs at two consecutive timepoints. The results showed that although the cartilage was still in a catabolic state directly after KJD, there were signs of healing at 10 weeks of follow-up. The signaling pathways involved in the healing process, including TGF-beta and Notch pathways, were explored.

Immune Responses and Pathogenesis following Experimental SARS-CoV-2 Infection in Domestic Cats

Summary: Several reports demonstrated domestic cats' susceptibility to SARS-CoV-2 infection. In this study, the immune responses in cats after experimental inoculation were investigated, along with infection kinetics and pathological lesions. Infected cats showed mild lung changes and viral antigen expression, but no clinical signs. Viral RNA was present in nasal and throat swabs until day 7, and infectious virus could be isolated from nose, trachea, and lungs until day 7. Cats developed a humoral immune response from day 7 onwards, with limited cellular immune responses on day 7.

Summary: This study systematically investigates the influence of O-antigen and Lipid A composition on the antibacterial and anti-endotoxin activity of four HDPs. The results show that O-antigen does not affect antibacterial activity, while Lipid A modification can affect the immunomodulatory role of HDPs. The findings have important implications for the application of HDPs in the treatment of infectious diseases.
