Summary: NPM1 is frequently overexpressed in colorectal cancer (CRC) and correlates with prognosis and sensitivity to chemotherapy. Inhibition of NPM1 enhances chemosensitivity of CRC cells through AKT pathway-mediated apoptosis. Suppressing NPM1 can attenuate tumor growth and restore chemosensitivity in CRC cells.

Summary: This study provides a comprehensive analysis of CoV-specific antibody repertoires in children and adults, revealing qualitative differences in antibody repertoires against endemic HCoVs between children and adults. Antibody specificities targeting the fusion peptide region and 52' cleavage site of the spike protein show broad cross-reactivity with peptides of epidemic human and nonhuman coronaviruses.

Summary: The study found a higher prevalence of herpesvirus infections in obese adults in Qatar, but this association was not observed in the pediatric population. There was a significant association between seropositivity to herpes simplex virus 1 (HSV-1) and obesity.

Summary: The study investigated a patient with HPV-2-driven "tree-man" phenotype and two relatives with severe HPV4-driven warts, revealing the relationship between a CD28 variant and T cell responses. Findings suggest that T cell responses are important in controlling HPV infections.

Summary: STK4 deficiency in patients can result in broad impairment of immune function beyond lymphoid cells, leading to recurrent viral and bacterial infections, abnormal T cell subsets, interferon signaling dysregulation, and distinctive antibody responses.

Summary: A study on a patient with a homozygous loss-of-function mutation in PDCD1 reveals dysregulated immune features involving IL-6, IL-23, STAT3, and ROR gamma T, which may be potential targets for the treatment of PD-1 blockade-induced autoimmunity. The research highlights the essential role of human PD-1 in governing antimycobacterial immunity and self-tolerance, while identifying actionable molecular targets for the diagnostic and therapeutic management of TB and autoimmunity in patients on PD-1 blockade.

Summary: This study explored the genetic variation of HLA class II genes in a cohort of Arab adults and found a strong association between zygosity of HLA class II genes and antibody responses to common pathogens. The results suggest that HLA class II gene polymorphisms confer specific humoral immunity against common pathogens, contributing to the genetic diversity of HLA class II loci during human evolution.

Summary: Inborn errors of IFN-g-dependent macrophagic immunity cause mycobacterial diseases, while inborn errors of IFN-a/b-dependent intrinsic immunity lead to viral diseases. Children with complete IRF1 deficiency have early-onset, life-threatening diseases caused by weakly virulent mycobacteria and related intramacrophagic pathogens. Their IFN-a/b-dependent antiviral immunity is largely normal, but their IFN-g-dependent macrophagic immunity is impaired.

Summary: Infectious diseases remain a major cause of morbidity and mortality worldwide, and metabolomics has the potential to serve as diagnostic biomarkers and enable personalized treatment. However, achieving personalized medicine requires collaboration across multiple disciplines, including technological development, bioscience, bioinformatics, biostatistics, clinicians, and biotechnology companies.
