Summary: This study reveals the important role of GLP-1R in the treatment of type 2 diabetes, and the rapid desensitization of stimulated GLP-1Rs by beta-arrestins. By studying KO mice, it was found that they had weaker responses to GLP-1R agonists, but showed improvement 6 hours after agonist injection. This study unveils fundamental aspects of GLP-1R response regulation with direct application to the rational design of GLP-1R-targeting therapeutics.

Summary: In this study, the effects of simvastatin on inflammatory and functional parameters of primary cortical and hypothalamic astrocyte cultures obtained from IFN alpha/beta receptor knockout (IFN alpha/beta R-/-) mice were investigated. The results showed that simvastatin modulated cytokine production and release, as well as the expression of cytoprotective genes and functional parameters in astrocytes from different brain regions.

Summary: Our study demonstrates that SELENOM knockout in mice results in decreased synaptic plasticity and memory impairment. Furthermore, SELENOM knockout leads to hyperglycemia and disrupts glucose metabolism, which is crucial for synapse formation and transmission in the brain. The inhibition of the brain insulin signaling pathway is identified as the underlying mechanism for the synaptic deficits and cognitive dysfunction observed in SELENOM knockout mice.

Summary: Research on Htr3a knockout mice revealed autistic-like behaviors, impaired memory, and reduced susceptibility to seizures. The upregulation of NMDAR in PV+ interneurons plays a critical role in regulating GABAergic input and may be involved in the pathogenesis of autism associated with HTR3A deficiency. The NMDAR system could be a potential therapeutic target for autism.

Summary: Recent studies have demonstrated that EPO exerts neuroprotective and neurotrophic effects on neurological disorders. However, the potential benefits of EPO on diabetic cognitive impairments have not been well studied. In this research, the authors investigated the effects of EPO on diabetic mice and HT22 cells. They found that EPO treatment improved cognitive impairments, lowered blood glucose levels, and alleviated damage in both the pancreas and hippocampus. EPO also regulated the expression of various factors related to cell survival and apoptosis in HT22 cells. The findings suggest that EPO plays a protective role in diabetic cognitive impairments by modulating BDNF and PSD-95 expression via NMDA receptors.
