Summary: N epsilon-lysine acetylation in the ER is crucial for quality control, with dysfunction of AT-1 leading to severe disease phenotypes. Two models of AT-1 dysregulation showed reorganization of the ER, ERGIC, and Golgi apparatus, highlighting the importance of AT-1 in maintaining proper organization and engagement of the secretory pathway.

Summary: The study found that disrupting Arc protein ubiquitination enhances DHPG-induced ER-mediated Ca2+ release. This alteration is observed in all neuronal subregions except secondary branchpoints. Disruptions in Arc ubiquitination increase Arc interaction with the integral ER protein Calnexin.

Summary: This study identified the accumulation of ER luminal proteins in the cytosol of glioblastoma cells, and demonstrated that ER protein reflux to the cytosol occurs upon ER proteostasis perturbation. The study also showed that refluxed, cytosolic AGR2 binds and inhibits the tumor suppressor p53, suggesting a mechanism named ER to Cytosol Signaling (ERCYS) which provides a selective advantage to tumor cells through gain-of-cytosolic functions.

Summary: This study reveals that cell stress induces the release of clusterin (CLU), a usually secreted chaperone, into the cytosol. CLU can interact with cytosolic misfolded proteins, directing them to degradation through autophagy and the proteasome, and protect cells from stress.

Summary: This study found that the endoplasmic reticulum protein ERp29 may act as a bridge linking the lectin chaperones calnexin and calreticulin, forming complexes such as CNX-CRT. This not only facilitates the binding of CNX and CRT, but also reveals a new function of ERp29.

Summary: The endoplasmic reticulum is equipped with protein disulfide isomerases, molecular chaperons, and other folding enzymes to ensure proper folding of newly synthesized proteins. Arabidopsis thaliana PDI11 interacts with ER resident lectin chaperones CRT1 and CRT2, with the D domain providing binding sites for this interaction. This implies that Arabidopsis CRT1/2 may specifically recruit AtPDI11 to assist in glycoprotein folding in the ER.

Summary: This study investigated how ER stress exposes the placenta to the risk of preeclampsia, revealing that CRT is upregulated in serum samples from preeclamptic women but not in placental specimens, and ER stress causes extracellular release of CRT, which suppresses cytotrophoblast syncytialization.

Summary: This study found that fenofibrate can alleviate ER stress and improve the pathology of NAFLD by regulating the expression of let-7 miRNA and SERCA2b. This provides new insights into the specific mechanism of fenofibrate in treating NAFLD and improving lipid metabolism disorders.

Summary: Muscle fiber is the basic unit of skeletal muscle and its type is closely related to meat quality. This study found that Mdfi can promote muscle fiber type transformation by activating CaMKK2 and AMPK phosphorylation, stimulating the conversion of C2C12 cells from glycolytic to oxidative type, and increasing mitochondrial biogenesis, aerobic metabolism, and calcium levels. These findings provide a better understanding of the regulatory mechanism of Mdfi in muscle fiber type transformation and suggest potential therapeutic targets for muscle and metabolic-related diseases.

Summary: Using single-cell and multilevel analyses, this study emphasizes the critical importance of the first exposure to calreticulin (CRT) in eliciting immunogenicity. The ERASION strategy, utilizing the high expression of functional proteins, including CRT, on the ER membrane, enables the targeting of tumor cells and immune effectors, promoting dendritic cell maturation and T cell infiltration, and eliciting an immunogenic effect from a nonimmunogenic chemotherapeutic drug.

Summary: This study identified a novel calcineurin regulatory-subunit-binding protein, MoCbp7, which is involved in calcium signal transduction and is important for the growth, conidiation, appressorium formation, invasive growth, and virulence of Magnaporthe oryzae. MoCbp7 also interacts with calcineurin to regulate endoplasmic reticulum homeostasis.

Summary: The study reveals that IP3R1 plays an important role in the maturation of pig oocytes by regulating the IP3R1-GRP75-VDAC1 channel between mitochondria and the endoplasmic reticulum (ER), affecting calcium balance. Inhibiting IP3R1 expression leads to calcium overload and mitochondrial oxidative stress, resulting in increased reactive oxygen species (ROS) levels and apoptosis. These findings have significant implications for obtaining high-quality eggs and maintaining preimplantation embryo development.
