Summary: This study reveals that alpha-ketoglutarate (AKG) can prevent and treat abdominal aortic aneurysm (AAA) through inhibiting oxidative stress and inflammation. AKG pre-treatment effectively prevents aneurysm dilation and aortic rupture, and reduces macrophage infiltration, elastin degradation, and collagen fibers remodeling. The study also demonstrates that AKG reduces the production of inflammatory cytokines, apoptosis, and excessive superoxide. Mechanistically, AKG regulates the PXDN/HOCL/ERK signaling pathway to exert its protective effects on AAA.

Through experiments using xenograft model and Ishikawa cells, the potential molecular mechanism of dioscin was explored. The results showed that dioscin inhibited Ishikawa cell migration and invasion through the MEK/ERK and JNK signaling pathways. Hub miRNAs and genes identified in this study could be potential molecular targets for future EC treatment.

Summary: This study examined the role of hepatoma-derived growth factor (HDGF) in gefitinib resistance in non-small cell lung cancer (NSCLC) and identified the underlying mechanisms. The results showed that HDGF overexpression aggravated the malignant phenotype of NSCLC cells, while HDGF knockdown reversed these effects. The overexpression of HDGF also led to gefitinib resistance, while knockdown enhanced gefitinib sensitivity. Higher levels of HDGF in plasma or tumor tissue were associated with gefitinib resistance. HDGF activated the Akt and ERK signaling pathways, independent of EGFR phosphorylation, contributing to gefitinib resistance.

Summary: This study investigated the effects of Bu-Gu-Sheng-Sui decoction (BGSSD) on osteogenesis and its potential mechanism. The results showed that BGSSD protected bone mass, increased the expression of osteogenic specific factor Runx2, and improved bone trabecular structure. In vitro, BGSSD accelerated the proliferation and differentiation of MC3T3-E1 cells and increased the expression of Runx2. Moreover, BGSSD effectively boosted the expression levels of ERK and Smad. These findings suggest that BGSSD may promote bone formation through the ERK/Smad pathways.

Summary: Recent research has identified a novel lncRNA Linc-KILH, which is significantly up-regulated in HCC tissues and positively correlated with tumor size, microvascular invasion, intrahepatic metastasis and decreased survival in HCC patients. Silencing Linc-KILH can inhibit the proliferation and metastasis abilities of KRT19-positive HCC cells. Mechanistically, Linc-KILH interacts with KRT19 and inhibits the phosphorylation of KRT19, leading to enhanced translocation of KRT19 in HCC cells.

Summary: This study evaluated the role of PDK4 in bladder cancer and its related protein changes. It was found that elevated expression of PDK4 was observed in high-grade bladder cancers. Knockdown of PDK4 in bladder cancer cells resulted in reduced cell migration and invasion. Additionally, a PDK4 knockdown xenograft model showed reduced bladder cancer growth. These findings suggest that PDK4 plays a critical role in the metastasis and growth of bladder cancer cells through changes in ERK, SRC, and JNK.
