Crystal Structure of the Dengue Virus Methyltransferase Bound to a 5′-Capped Octameric RNA

The N-terminal domain of the flavivirus NS5 protein functions as a methyltransferase (MTase). It sequentially methylates the N7 and 2'-O positions of the viral RNA cap structure (GpppA ->(7me)GpppA ->(7me)GpppA(2'-O- me)). The same NS5 domain could also have a guanylyltransferase activity (GTP+ppA-RNA -> GpppA). The mechanism by which this protein domain catalyzes these three distinct functions is currently unknown. Here we report the crystallographic structure of DENV-3 MTase in complex with a 5'-capped RNA octamer (G(ppp)AGAACCUG) at a resolution of 2.9 angstrom. Two RNA octamers arranged as kissing loops are encircled by four MTase monomers around a 2-fold non-crystallography symmetry axis. Only two of the four monomers make direct contact with the 5' end of RNA. The RNA structure is stabilised by the formation of several intra and intermolecular base stacking and non- canonical base pairs. The structure may represent the product of guanylylation of the viral genome prior to the subsequent methylation events that require repositioning of the RNA substrate to reach to the methyl-donor sites. The crystal structure provides a structural explanation for the observed trans-complementation of MTases with different methylation defects.

Summary: Alphaviruses are mosquito-borne pathogens that can cause arthritis or encephalitis. The nonstructural protein 4 (nsP4) of alphaviruses plays a crucial role in viral RNA replication. The crystal structures of the RdRp domain of nsP4 from Ross River virus and Sindbis virus were determined and found to be closely related to RdRps from other viruses. Understanding the structure and function of nsP4 is important for developing antiviral drugs.

Targeting SARS-CoV-2 and host cell receptor interactions

Summary: Despite the availability of vaccines and therapeutics, continuous genetic alterations in the SARS-CoV-2 virus pose a persistent threat, especially to immunocompromised and elderly individuals. The virus enters host cells via the interaction of its spike protein (S protein) with different receptors on the cell surface, such as angiotensin-converting enzyme 2 (ACE2). This review discusses therapeutic approaches to block the interaction between SARS-CoV-2 and host cell receptors, as well as the identification of auxiliary entry receptors.

Molecular architecture of the Chikungunya virus replication complex

Summary: This study reveals the high-resolution structure of the core RNA replicase of chikungunya virus and the native RC architecture in its cellular context using in vitro reconstitution and in situ electron cryotomography. The viral polymerase nsP4, in complex with nsP2 helicase-protease, is located in the central pore of the membrane-anchored nsP1 RNA-capping ring within the core RNA replicase. Additionally, a large cytoplasmic ring is found next to the C terminus of nsP1, forming the holo-RNA-RC at the neck of spherules formed in virus-infected cells.
