Inflammatory Mediator TAK1 Regulates Hair Follicle Morphogenesis and Anagen Induction Shown by Using Keratinocyte-Specific TAK1-Deficient Mice

Transforming growth factor-beta-activated kinase 1 (TAK1) is a member of the NF-kappa B pathway and regulates inflammatory responses. We previously showed that TAK1 also regulates keratinocyte growth, differentiation, and apoptosis. However, it is unknown whether TAK1 has any role in epithelial-mesenchymal interactions. To examine this possibility, we studied the role of TAK1 in mouse hair follicle development and cycling as an instructive model system. By comparing keratinocyte-specific TAK1-deficient mice (Map3k7(fl/fl)K5-Cre) with control mice, we found that the number of hair germs (hair follicles precursors) in Map3k7(fl/fl)K5-Cre mice was significantly reduced at E15.5, and that subsequent hair follicle morphogenesis was retarded. Next, we analyzed the role of TAK1 in the cyclic remodeling in follicles by analyzing hair cycle progression in mice with a tamoxifen-inducible keratinocyte-specific TAK1 deficiency (Map3k7(fl/fl)K14-Cre-ER(T2)). After active hair growth (anagen) was induced by depilation, TAK1 was deleted by topical tamoxifen application. This resulted in significantly retarded anagen development in TAK1-deficient mice. Deletion of TAK1 in hair follicles that were already in anagen induced premature, apoptosis-driven hair follicle regression, along with hair follicle damage. These studies provide the first evidence that the inflammatory mediator TAK1 regulates hair follicle induction and morphogenesis, and is required for anagen induction and anagen maintenance.

Targeting mitochondria in dermatological therapy: beyond oxidative damage and skin aging

Summary: The analysis of mitochondria in oxidative damage and skin aging is important in dermatological research. Mitochondria produce reactive oxygen species (ROS) that can be cytotoxic and DNA-damaging, leading to (photo-)aging. However, mitochondria also have physiological functions and their dysfunction is involved in skin diseases and cancers. This paper explores the role of mitochondria in skin physiology and evaluates potential lead compounds for therapeutic research. The mitochondrial protein MPZL3 is highlighted as a promising target for future research.

Concentration-dependent stimulation of melanin production as well as melanocyte and keratinocyte proliferation by melatonin in human eyelid epidermis

Summary: This pilot study aimed to investigate the effects of melatonin on melanin production and melanocytes in intact human epidermis. The results showed that melanin content and melanocyte dendricity increased with melatonin treatment, while tyrosinase activity was inhibited. Higher doses of melatonin increased the number and proliferation of melanocytes and keratinocytes, as well as the expression of the premelanosomal marker gp100. These findings suggest that long-term melatonin therapy may stimulate melanogenesis and increase the activity of epidermal melanocytes.

Hypoxia-inducible factor 1A inhibition overcomes castration resistance of prostate tumors

Summary: Androgen deprivation therapy is a key treatment for prostate cancer, but some cases develop into castration-resistant prostate cancer (CRPC). In this study, it was found that the major luminal cell population in CRPC is castration-resistant, with enhanced expression of inflammation and stemness markers. Additionally, hypoxia-inducible factor 1 (HIF1) signaling, which promotes malignant progression, is further activated. Inhibition of HIF1A enhances the sensitivity of Pten-deficient prostatic tumors to castration and induces apoptotic signaling in human CRPC cell lines.

Preliminary evidence that Merkel cells exert chemosensory functions in human epidermis

Summary: Recent research has found that epidermal Merkel cells (MCs) not only have mechanotransduction function for light-touch sensory stimuli, but also are thermosensitive, suggesting a physiological role beyond mechanosensation. Functional olfactory receptors, specifically OR2AT4, were found to be expressed by epidermal MCs in healthy human skin, similar to neighboring keratinocytes. Stimulation of OR2AT4 by topical application of the synthetic odorant Sandalore significantly increased Piccolo protein expression in MCs, indicating increased vesicle trafficking and recycling, and reduced nerve growth factor (NGF) immunoreactivity within MCs, potentially suggesting increased neurotrophin release upon OR2AT4 activation. Further investigation is needed to explore whether these newly discovered properties of MCs are dysregulated in specific skin disorders and if they can be targeted therapeutically to maintain/promote skin health.
