Background: Increased prevalence of atherosclerotic cardiovascular disease in HIV-infected patients has been observed. The cause of this accelerated atherosclerosis is a matter of controversy. As clinical studies are complicated by a multiplicity of risk-factors and a low incidence of hard endpoints, studies in animal models could be attractive alternatives. Methodology/Principal Findings: We evaluated gene expression of lectin-like oxidized-low-density-lipoprotein receptor-1 (LOX-1), vascular cell adhesion molecule-1 (VCAM-1), and intercellular adhesion molecule-1 ( ICAM- 1) in HIV-1 transgenic (HIV-1Tg) rats; these genes are all thought to play important roles in early atherogenesis. Furthermore, the plasma level of sICAM-1 was measured. We found that gene expressions of LOX-1 and VCAM- 1 were higher in the aortic arch of HIV-1Tg rats compared to controls. Also, the level of sICAM-1 was elevated in the HIV-1Tg rats compared to controls, but the ICAM- 1 gene expression profile did not show any differences between the groups. Conclusions/Significance: HIV-1Tg rats have gene expression patterns indicating endothelial dysfunction and accelerated atherosclerosis in aorta, suggesting that HIV-infection per se may cause atherosclerosis. This transgenic rat model may be a very promising model for further studies of the pathophysiology behind HIV-associated cardiovascular disease.

Synthesis and in vivo evaluation of [11C]tucatinib for HER2-targeted PET imaging

Marius Mueller, Vladimir Shalgunov, Lars Hvass, Jesper T. Jorgensen, Vasko Kramer, Markus Staudt, Umberto Maria Battisti, Andreas Kjaer, Matthias M. Herth

Summary: This article aims to develop the first small molecule HER2 PET imaging probe based on tucatinib that can penetrate the blood-brain barrier. Despite its high affinity, no significant tumor accumulation was observed for [11C]tucatinib, possibly due to its high lipophilicity. Chemical modifications are required to increase the polarity of [11C]tucatinib for tumor imaging.

Synthesis and radiolabeling of a polar [125I]I-1,2,4,5-tetrazine

Natasha Bidesi, Vladimir Shalgunov, Umberto Maria Battisti, Lars Hvass, Jesper Tranekjaer Jorgensen, Christian B. M. Poulie, Andreas I. Jensen, Andreas Kjaer, Matthias M. Herth

Summary: Pretargeting imaging has gained attention for its excellent bioorthogonality and improved imaging contrast compared to conventional imaging. A new iodo tetrazine derivative has been synthesized and developed into an iodine-125 analog. Radiolabeling with N-chlorosuccinimide or chloramine-T in MeCN or MeOH resulted in a radiochemical conversion of >80%. Deprotection was successful when the radiolabeled tetrazine was stirred in 6-M HCl(aq.) at 60 degrees C for 2.5 h. This is the first iodine-labeled H-tetrazine. In vivo investigations on its pretargeting ability are currently underway.

Eva Lykke, Anders Christensen, Karina Juhl, Ulla Feldt-Rasmussen, Mette Friberg Hitz, Sannia Mia Svenningsen Sjostedt, Christoffer Holst Hahn, Ditte Maria Kraik Svensson, Karoline Kanstrup Springborg, Mads Georg Stage, Gitte Bjorn Hvilsom, Linda Maria Hilsted, Morten Dahl, Giedrius Lelkaitis, Andreas Kjaer, Preben Homoe, Christian von Buchwald

Summary: The purpose of this study was to investigate whether near-infrared autofluorescence image guidance can reduce the incidence of hypoparathyroidism in total thyroidectomy. The results showed that near-infrared autofluorescence imaging can improve the detection rate of parathyroid glands and reduce the rate of hypoparathyroidism, but the difference was not statistically significant. Further studies are needed to confirm the use of near-infrared autofluorescence imaging as an auxiliary tool for parathyroid gland identification.

Subdural Levels of Interleukin 1-receptor Antagonist are Elevated in Patients with Recurrent Chronic Subdural Hematomas

Thorbjorn Soren Ronn Jensen, Tina Binderup, Markus Harboe Olsen, Andreas Kjaer, Kare Fugleholm

Summary: Anti-inflammatory treatment reduces the risk of recurrent chronic subdural hematoma (CSDH), but clinical implementation is improper due to side effects. This study aims to investigate the associations between cytokine levels and the risk of recurrence in CSDH patients and identify potential medical targets. The study found elevated levels of interleukin-1 receptor antagonist (IL-1ra) in recurrent CSDH patients, suggesting IL-1ra as a possible target for medical treatment.

Propagation Capacity of Phage Display Peptide Libraries Is Affected by the Length and Conformation of Displayed Peptide

Danna Kamstrup Sell, Anders Wilgaard Sinkjaer, Babak Bakhshinejad, Andreas Kjaer

Summary: Mixing multiple peptide libraries with different displayed peptide characteristics can increase the throughput of selection. This study used next-generation sequencing to investigate the amplification capacity of three widely used commercial phage display peptide libraries and found that the length and cyclic conformation of displayed peptides can affect the propagation capacity of the libraries. Additionally, the Ph.D.(TM)-7 library showed higher propagation capacity. These findings provide evidence for considering peptide characteristics when pooling multiple combinatorial libraries for phage display selection.
