miR-155 Inhibition Sensitizes CD4+Th Cells for TREG Mediated Suppression

Background: In humans and mice naturally occurring CD4(+)CD25(+) regulatory T cells (nTregs) are a thymus-derived subset of T cells, crucial for the maintenance of peripheral tolerance by controlling not only potentially autoreactive T cells but virtually all cells of the adaptive and innate immune system. Recent work using Dicer-deficient mice irrevocably demonstrated the importance of miRNAs for nTreg cell-mediated tolerance. Principal Findings: DNA-Microarray analyses of human as well as murine conventional CD4(+) Th cells and nTregs revealed a strong up-regulation of mature miR-155 (microRNA-155) upon activation in both populations. Studying miR-155 expression in FoxP3-deficient scurfy mice and performing FoxP3 ChIP-Seq experiments using activated human T lymphocytes, we show that the expression and maturation of miR-155 seem to be not necessarily regulated by FoxP3. In order to address the functional relevance of elevated miR-155 levels, we transfected miR-155 inhibitors or mature miR-155 RNAs into freshly-isolated human and mouse primary CD4(+) Th cells and nTregs and investigated the resulting phenotype in nTreg suppression assays. Whereas miR-155 inhibition in conventional CD4(+) Th cells strengthened nTreg cell-mediated suppression, overexpression of mature miR-155 rendered these cells unresponsive to nTreg cell-mediated suppression. Conclusion: Investigation of FoxP3 downstream targets, certainly of bound and regulated miRNAs revealed the associated function between the master regulator FoxP3 and miRNAs as regulators itself. miR-155 is shown to be crucially involved in nTreg cell mediated tolerance by regulating the susceptibility of conventional human as well as murine CD4(+) Th cells to nTreg cell-mediated suppression.

Summary: This study generated a mouse model with a knockout of the CD18 gene specifically in dendritic cells, and found that it affected cytokine production, signal transduction, and inflammatory signaling. In a mouse model of experimental autoimmune encephalomyelitis, the knockdown of beta 2 integrins in dendritic cells resulted in a delayed onset and milder course of the disease, associated with lower frequencies of Th1/Th17 cells in the spinal cord.

This mini-review discusses the role of LFA-1 in Treg induction, maintenance, and regulatory function using various beta(2)-integrin-deficient mouse models.

Summary: The analysis of secretome provides important information on intercellular communication and cell behavior in specific tissues. Especially in tumors, secretome data can aid in diagnosis and therapy decisions. Mass spectrometry-based analysis of cell-conditioned media is commonly used for unbiased characterization of cancer secretomes in vitro. Metabolic labeling using azide-containing amino acid analogs facilitates this analysis in the presence of serum. However, these modified analogs may affect protein folding and incorporation into newly synthesized proteins. Combining transcriptome and proteome analysis, we investigate the effects of azidohomoalanine labeling on gene and protein expression. Our findings indicate that AHA labeling induces changes in transcript and protein expression, leading to cellular stress, apoptosis-related pathways, and alteration of secretome composition on a global scale.

NFATc1 induction by an intronic enhancer restricts NKT yδ cell formation

Summary: In the thymus, the deletion of NFATc1 or its inducible isoforms during the DN stages of thymocyte development leads to an increase in gamma delta thymocytes while alpha beta thymocytes are mostly unaffected. These gamma delta thymocytes upregulate PLZF, the master regulator of NKT cell development, acquiring an NKT gamma delta cell phenotype with higher survival rates. The enhancer E2 is crucial for NFATc1's suppressive function in NKT gamma delta cell formation, translating a strong gamma delta TCR signal into inducible expression of NFATc1 isoforms to control the numbers of NKT gamma delta cells.

Assessing the immune response to SARS-CoV-2 mRNA vaccines in siponimod-treated patients: a nonrandomized controlled clinical trial (AMA-VACC)
