A Method to Quantify Mouse Coat-Color Proportions

Coat-color proportions and patterns in mice are used as assays for many processes such as transgene expression, chimerism, and epigenetics. In many studies, coat-color readouts are estimated from subjective scoring of individual mice. Here we show a method by which mouse coat color is quantified as the proportion of coat shown in one or more digital images. We use the yellow-agouti mouse model of epigenetic variegation to demonstrate this method. We apply this method to live mice using a conventional digital camera for data collection. We use a raster graphics editing program to convert agouti regions of the coat to a standard, uniform, brown color and the yellow regions of the coat to a standard, uniform, yellow color. We use a second program to quantify the proportions of these standard colors. This method provides quantification that relates directly to the visual appearance of the live animal. It also provides an objective analysis with a traceable record, and it should allow for precise comparisons of mouse coats and mouse cohorts within and between studies.

Tracking evoked responses to auditory and visual stimuli in fetuses exposed to maternal high-risk conditions

Summary: This study tracked the development trajectory of fetal auditory and visual evoked responses in high-risk pregnancies, showing a decrease in latency with gestational age progression. There was a significant difference in detection rates between male and female fetuses, but this difference was not reflected in latency values or sensory modality. Factors affecting response detectability included intervening layers of adipose tissue and an increase in maternal body mass index.

Narcotic Refills and Patient Satisfaction With Pain Control After Total Joint Arthroplasty

Summary: This study reviewed preoperative and postoperative narcotic prescriptions for 438 consecutive TJA patients using the state's mandatory opioid prescription monitoring program. Patients who underwent total hip arthroplasty, were aged 65 or older, and had Medicare coverage were more likely to receive fewer post-discharge narcotic refills. Patients with higher satisfaction tended to consume less overall opioids when needing a refill.

Lymph Liquid Biopsy for Detection of Cancer Stem Cells

Summary: Blood liquid biopsy is commonly used to detect markers of metastatic tumors, but the utility of lymphatic markers remains largely unexplored. This study introduces lymph liquid biopsy to detect stem-like CTCs in lymph samples, highlighting the significance of the lymphatic pathway in metastasis development. Integration of lymph and blood liquid biopsies showed promise for diagnosis and prognosis of early metastasis.

Transfers of pediatric patients with isolated injuries to a rural Level 1 Orthopedic Trauma Center in the United States: are they all necessary?

Summary: Approximately 14.6% of patients transferred to a pediatric Level 1 trauma center for isolated orthopedic injuries underwent no surgery or fracture reductions and were discharged directly home. In particular, isolated tibia fractures were more frequently treated without reduction or surgery. Telemedicine consultation for these specific injury types may limit unnecessary and costly transfers to a Level 1 pediatric trauma hospital in the future.

Spectroscopic investigation of radiation-induced reoxygenation in radiation-resistant tumors

Summary: In this study, changes in hemoglobin oxygen saturation (sO(2)) in tumor xenografts from two head and neck squamous cell carcinoma cell lines during radiation therapy were monitored. The sO(2) in treatment-responsive UM-SCC-22B tumors immediately increased post-radiation, while in the relatively radiation-resistant UM-SCC-47 tumors, sO(2) increased after each radiation dose, possibly due to HIF-1 expression.

Evaluation of Pelvic Floor Muscles in Pregnancy and Postpartum With Non-Invasive Magnetomyography

Summary: This study used a non-invasive and novel magnetomyography (MMG) technique to record and characterize the activity of levator ani muscles (LAM) during pregnancy and postpartum. The results showed changes in amplitude and spectral parameters of LAM activity during postpartum. This study provides a new method for studying the changes in LAM activity during pregnancy and postpartum.

VT68.2: An Antibody to Chondroitin Sulfate Proteoglycan 4 (CSPG4) Displays Reactivity against a Tumor-Associated Carbohydrate Antigen

Summary: The study demonstrated that anti-CSPG4 monoclonal antibodies exhibit anti-tumor activity and therapeutic potential for breast cancer treatment. The specificity and importance of glycan contribution to the binding of specific anti-CSPG4 antibodies to tumor cells were investigated. The mAb VT68.2 displayed strong binding to a carbohydrate-mimicking peptide (CMP) and showed a higher affinity for certain glycans. Furthermore, VT68.2 inhibited cancer cell migration, suggesting its potential as a less toxic therapeutic approach targeting tumor-specific glyco-peptides.

The effect of maternal pregestational diabetes on fetal autonomic nervous system

Summary: The study found that maternal pregestational diabetes has an effect on the fetal autonomic nervous system (ANS), which can be assessed using non-invasive fetal magnetocardiography. There were differences in the frequency domain of fetal heart rate variability (fHRV) and heart rate-movement coupling in fetuses of pregestational diabetic mothers compared to non-diabetics. However, the effect on fHRV related to fetal ANS and sympathovagal balance was not as conclusive as observed in neonates of pregestational diabetic mothers.
