Negative Smad Expression and Regulation in the Developing Chick Limb

The inhibitory or negative Smads, Smad6 and Smad7, block TGF beta superfamily signals of both the BMP and TGF beta classes by antagonizing the intracellular signal transduction machinery. We report the cloning of one Smad6 and two Smad7 (Smad7a and Smad7b) chick homologs and their expression and regulation in the developing limb. Smad6 and Smad7a are expressed in dynamic patterns reflecting the domains of BMP gene expression in the limb. Activation and inhibition of the BMP signaling pathway in limb mesenchyme indicates that negative Smad gene expression is regulated, at least in part, by BMP family signals.

Recent research on thalidomide analogues shows promise in retaining clinical benefits without harmful effects. Understanding the challenges faced by thalidomide survivors as they age can improve healthcare for them and other individuals with congenital limb differences.

A New Generation of IMiDs as Treatments for Neuroinflammatory and Neurodegenerative Disorders

Summary: The immunomodulatory imide drug (IMiD) class, consisting of thalidomide and its derivatives lenalidomide and pomalidomide, has greatly improved the treatment of specific cancers and possesses potent anticancer and anti-inflammatory actions. These actions are mainly mediated by the binding of IMiDs to the human protein cereblon, resulting in the modification of protein degradation and the emergence of new substrate proteins. However, this binding also leads to adverse effects, including teratogenicity.

Summary: Despite whole-genome sequencing (WGS), many single-gene disorders remain unsolved. To address this issue, researchers have developed an analytic tool called GROFFFY, which integrates coordinates for regions with experimental evidence of functionality and can prioritize variants in non-coding regions. Applied to WGS data, GROFFFY reduced the number of variants without deleting disease-causal variants. It successfully identified rare deletions in the 3' untranslated region of the tumor suppressor gene SMAD4, shedding light on the impact of these variants on regulatory systems based on RNA polyadenylation.
