MGluR5 Mediates the Interaction between Late-LTP, Network Activity, and Learning

Hippocampal synaptic plasticity and learning are strongly regulated by metabotropic glutamate receptors (mGluRs) and particularly by mGluR5. Here, we investigated the mechanisms underlying mGluR5-modulation of these phenomena. Prolonged pharmacological blockade of mGluR5 with MPEP produced a profound impairment of spatial memory. Effects were associated with 1) a reduction of mGluR1a-expression in the dentate gyrus; 2) impaired dentate gyrus LTP; 3) enhanced CA1-LTP and 4) suppressed theta (5-10 Hz) and gamma (30-100 Hz) oscillations in the dentate gyrus. Allosteric potentiation of mGluR1 after mGluR5 blockade significantly ameliorated dentate gyrus LTP, as well as suppression of gamma oscillatory activity. CA3-lesioning prevented MPEP effects on CA1-LTP, suggesting that plasticity levels in CA1 are driven by mGluR5-dependent synaptic and network activity in the dentate gyrus. These data support the hypothesis that prolonged mGluR5-inactivation causes altered hippocampal LTP levels and network activity, which is mediated in part by impaired mGluR1-expression in the dentate gyrus. The consequence is impairment of long-term learning.

Postweaning social isolation and autism-like phenotype: A biochemical and behavioral comparative analysis

Summary: Adolescence is a critical period for brain development, and disturbances during this time can increase the risk of neuropsychiatric disorders. This study compared the effects of postweaning social isolation (PWSI) on inbred C57BL/6 N mice and BTBR mice, which are a model for autism spectrum disorders. The results showed that both PWSI mice and BTBR mice exhibited reduced social responsiveness and altered levels of certain receptors in the brain, suggesting that absence of social stimuli during adolescence can induce a phenotype similar to autism spectrum disorders.

Summary: Breakthrough cancer pain (BTcP) is a sudden and transient exacerbation of pain in patients treated with opioid analgesics. Light-activated drugs offer a novel strategy for rapid pain control without systemic side effects. Activation of mGlu5 receptors in the thalamus with light-induced compounds showed analgesic effects in inflammatory and neuropathic pain models. This study demonstrates the potential use of optopharmacology in BTcP treatment.

Characterisation of the neural basis underlying appetitive extinction & renewal in Cacna1c rats

Summary: Recent studies have found that Cacna1c +/- heterozygous animals with impaired Cav 1.2 L-type voltage-gated calcium channels show difficulties in aversive forms of learning, but the role of these channels in extinction of appetitive associations is understudied. In this study, Cacna1c +/- male rats and their wild-type littermates were evaluated in an appetitive Pavlovian conditioning task to examine extinction learning and associated changes in the brain. The results showed that Cacna1c +/- animals adapted their responses during appetitive extinction and renewal, with distinct neural activation patterns observed in different brain regions. These findings provide novel evidence of brain plasticity in Cacna1c +/- rats after appetitive extinction and renewal. This study contributes to understanding the role of Cav 1.2 L-type voltage-gated calcium channels in neuropsychiatric disorders.

Summary: Task-free functional connectivity in animal models is an important tool for studying connectivity phenomena. However, the lack of standardized protocols and analysis methods hampers result comparison and integration. In this study, the authors developed a standardized protocol, called StandardRat, for rat functional magnetic resonance imaging (fMRI) acquisition and analysis, which enhances the detection of functional connectivity patterns and promotes cooperation in neuroscience research.

Oscillatory Deficits in the Sub-Chronic PCP Rat Model for Schizophrenia Are Reversed by mGlu5 Receptor-Positive Allosteric Modulators VU0409551 and VU0360172

Summary: The cognitive deficits of schizophrenia are caused by imbalanced excitatory and inhibitory signalling in the prefrontal cortex (PFC), leading to disrupted gamma oscillations. Two mGlu5 receptor-positive allosteric modulators (PAMs), VU0409551 and VU0360172, have been shown to restore cognitive deficits in a rodent model of schizophrenia by targeting PFC intracellular signalling molecules. In this study, gamma oscillatory activity was assessed in PFC slices from the rodent model, and the effects of the two PAMs were investigated. The results demonstrated that both VU0409551 and VU0360172 rescued the oscillatory deficits in a concentration-dependent manner, with the involvement of mGlu5 receptor modulation, PLC inhibition, and PKC inhibition.

The Metabotropic Glutamate 5 Receptor in Sleep and Wakefulness: Focus on the Cortico-Thalamo-Cortical Oscillations

Summary: Sleep is a complex behavior found in all animals, but our understanding of the neural circuits regulating sleep and wakefulness is limited. This review focuses on the role of mGlu5 receptors in the thalamocortical circuitry during normal and pathological sleep states. It also explores the potential of targeting mGlu5 receptors as a therapeutic approach for sleep disorders associated with epileptic seizures.
