Characterization of Plant Carotenoid Cyclases as Members of the Flavoprotein Family Functioning with No Net Redox Change

The later steps of carotenoid biosynthesis involve the formation of cyclic carotenoids. The reaction is catalyzed by lycopene beta-cyclase (LCY-B), which converts lycopene into beta-carotene, and by capsanthin-capsorubin synthase (CCS), which is mainly dedicated to the synthesis of kappa-cyclic carotenoids (capsanthin and capsorubin) but also has LCY-B activity. Although the peptide sequences of plant LCY-Bs and CCS contain a putative dinucleotide-binding motif, it is believed that these two carotenoid cyclases proceed via protic activation and stabilization of resulting carbocation intermediates. Using pepper (Capsicum annuum) CCS as a prototypic carotenoid cyclase, we show that the monomeric protein contains one noncovalently bound flavin adenine dinucleotide (FAD) that is essential for enzyme activity only in the presence of NADPH, which functions as the FAD reductant. The reaction proceeds without transfer of hydrogen from the dinucleotide cofactors to beta-carotene or capsanthin. Using site-directed mutagenesis, amino acids potentially involved in the protic activation were identified. Substitutions of alanine, lysine, and arginine for glutamate-295 in the conserved 293-FLEET-297 motif of pepper CCS or LCY-B abolish the formation of beta-carotene and kappa-cyclic carotenoids. We also found that mutations of the equivalent glutamate-196 located in the 194-LIEDT-198 domain of structurally divergent bacterial LCY-B abolish the formation of beta-carotene. The data herein reveal plant carotenoid cyclases to be novel enzymes that combine characteristics of non-metal-assisted terpene cyclases with those attributes typically found in flavoenzymes that catalyze reactions, with no net redox, such as type 2 isopentenyl diphosphate isomerase. Thus, FAD in its reduced form could be implicated in the stabilization of the carbocation intermediate.

Effect of lignocellulosic composition of Reutealis trisperma waste on nanocrystalline cellulose properties

Summary: This study investigated the influence of lignocellulosic composition on the characteristics of nanocrystalline cellulose (NCs) in different segments of Reutealis trisperma waste. The concentration of lignin, hemicellulose, and cellulose determined the properties of isolated NCs. The findings suggest that the different segments of Reutealis trisperma waste produce NCs with varying characteristics, including morphology, thermal stability, and size.

Summary: Natural products have gained attention in recent years due to rising healthcare expenses and global financial crises. Tamanu seed oil, a traditional remedy and cosmetic ingredient, contains coumarins, a group of phenolics with various beneficial properties. This study focused on the extraction of coumarin mixture from tamanu oil using ethanol, which resulted in high purity and recovery rates. Furthermore, other compounds were also detected in the polar fraction of tamanu oil.

Lutein Decreases Inflammation and Oxidative Stress and Prevents Iron Accumulation and Lipid Peroxidation at Glutamate-Induced Neurotoxicity

Summary: The xanthophyll carotenoid lutein has been shown to have protective effects in light-induced oxidative stress and may also play a neuroprotective role. Glutamate, a major neurotransmitter, can cause oxidative stress and neurotoxicity. This study demonstrates that glutamate affects oxidative stress, inflammation, iron metabolism, and lipid peroxidation in neuroblastoma cells, while lutein attenuates these effects. These findings suggest that lutein could be a beneficial treatment in neurodegenerative disorders.

SlIAA9 Mutation Maintains Photosynthetic Capabilities under Heat-Stress Conditions

Summary: Tomato is highly sensitive to temperature changes and climate change-induced global warming poses a major threat to its production. In this study, the photosynthetic capability of iaa9-3 and iaa9-5 mutants under heat-stress conditions was examined. It was found that these mutants maintained their photosynthetic capability even after 14 days of heat treatment, indicating the beneficial effect of the SlIAA9 mutation on plant adaptation to heat stress.

Parthenocarpic tomato mutants, iaa9-3 and iaa9-5, show plant adaptability and fruiting ability under heat-stress conditions

Summary: Fruit set is a major issue in tomato plants under heat-stress conditions, but parthenocarpic tomatoes can increase productivity in such conditions. This study aimed to assess the adaptability and fruiting capability of iaa9-3 and iaa9-5 tomato mutants under heat-stress conditions. The mutants showed delayed flowering, increased flower number, and improved fruit set and quality under heat stress. They also exhibited lower electrolyte leakage and H2O2 concentration, as well as higher antioxidant activities compared to the wild-type. These results suggest that iaa9-3 and iaa9-5 could be valuable genetic resources for heat-tolerant tomato development.

Summary: This study aimed to determine the full-length transcript of the genes of three types of tomatoes using long-reads sequencing technology. The results showed that there were 965 genes expressed in the iaa9-3 mutant, 2332 genes expressed in the iaa9-5 mutant, and 1536 genes specifically expressed in the wild type. Cluster analysis revealed distinct differences in transcript levels between the wild type and the mutants.

Plastid DNA Barcoding and RtActin cDNA Fragment Isolation of Reutealis Trisperma: A Promising Bioresource for Biodiesel Production

Summary: This study aimed to evaluate the application of plastid loci, rbcL and matK, as a DNA barcode for R trisperma, and to isolate and clone the RtActin (RtACT) gene fragment for gene expression studies. The 592 and 840 bp RtrbcL and RtmatK fragment genes were successfully isolated and cloned, and phylogenetic analysis showed that R trisperma is closely related to the Vernicia fordii Actin gene. The results suggest that RtrbcL could be further developed as a barcoding marker for R trisperma, and the RtACT gene could be investigated for gene expression studies in plants.
