P>Vacuolar sorting receptors (VSRs) are type-I integral membrane proteins that mediate biosynthetic protein traffic in the secretory pathway to the vacuole, whereas secretory carrier membrane proteins (SCAMPs) are type-IV membrane proteins localizing to the plasma membrane and early endosome (EE) or trans-Golgi network (TGN) in the plant endocytic pathway. As pollen tube growth is an extremely polarized and highly dynamic process, with intense anterograde and retrograde membrane trafficking, we have studied the dynamics and functional roles of VSR and SCAMP in pollen tube growth using lily (Lilium longiflorum) pollen as a model. Using newly cloned lily VSR and SCAMP cDNA (termed LIVSR and LISCAMP, respectively), as well as specific antibodies against VSR and SCAMP1 as tools, we have demonstrated that in growing lily pollen tubes: (i) transiently expressed GFP-VSR/GFP-LIVSR is located throughout the pollen tubes, excepting the apical clear-zone region, whereas GFP-LISCAMP is mainly concentrated in the tip region; (ii) VSRs are localized to the multivesicular body (MVB) and vacuole, whereas SCAMPs are localized to apical endocytic vesicles, TGN and vacuole; and (iii) microinjection of VSR or SCAMP antibodies and LlVSR small interfering RNAs (siRNAs) significantly reduced the growth rate of the lily pollen tubes. Taken together, both VSR and SCAMP are required for pollen tube growth, probably working together in regulating protein trafficking in the secretory and endocytic pathways, which need to be coordinated in order to support pollen tube elongation.

Summary: This study confirms the important role of JcDGAT1 and JcDGAT2 in regulating oil production in Jatropha curcas. Additionally, transgenic plants showed a significant increase in leaf TAG content, indicating the substrate specificity of JcDGAT1 and JcDGAT2 in seeds and leaves.

Genome-Wide Identification and Expression Analysis of Cytokinin Response Regulator (RR) Genes in the Woody Plant Jatropha curcas and Functional Analysis of JcRR12 in Arabidopsis

Summary: The cytokinin response regulator gene family plays a crucial role in regulating plant development and environmental responses. This study focused on the role of CK signaling in the growth and development of Jatropha curcas. Through bioinformatics and expression analysis, the CK response regulator genes in J. curcas were characterized, revealing their involvement in plant hormone, light, and abiotic stress responses. Evolutionary analysis and genomic comparison provided insights into the expansion mechanism of this gene family. RNA sequencing and quantitative reverse transcription-polymerase chain reaction were used to study the expression patterns of these genes in different tissues and their response to hormones and stress. Functional validation in Arabidopsis thaliana confirmed the importance of CK response regulator genes in growth and development processes.

Transcriptome-Based Construction of the Gibberellin Metabolism and Signaling Pathways in Eucalyptus grandis x E. urophylla, and Functional Characterization of GA20ox and GA2ox in Regulating Plant Development and Abiotic Stress Adaptations

Summary: This study identified 59,948 expressed genes from major vegetative tissues of Eucalyptus through transcriptome sequencing. Overexpression of EguGA20ox1 and EguGA20ox2 in Arabidopsis promoted vegetative growth but increased sensitivity to stress, while EguGA2ox1 overexpression enhanced stress resistance. Overexpression of EguGA20ox in Eucalyptus roots accelerated hairy root growth and improved xylem differentiation. This study provides comprehensive insights into GA metabolism and signaling genes in Eucalyptus and identifies their role in growth, stress tolerance, and xylem development, benefiting molecular breeding efforts.

Spatiotemporal organization and correlation of tip-focused exocytosis and endocytosis in regulating pollen tube tip growth

Summary: Using fluorescence recovery after photobleaching, we observed the occurrence and localization of rapid vesicular activities in the tip region of growing pollen tube. We demonstrated that both vesicle polar exocytic fusion and endocytosis take place at the apex of the pollen tube. Furthermore, we found that the dynamic activities of tip-focused exocytosis and endocytosis are closely correlated with the spatial organization of actin fringe.
