Phylogenetic and Biological Characterization of Fusarium oxysporum Isolates Associated with Onion in South Africa

Fusarium oxysporum f. sp. cepae causes Fusarium basal rot of onion, a disease of worldwide importance. Limited information is available on the phylogenetic diversity, vegetative compatibility groups (VCGs), mating type idiomorphs, and virulence of F oxysporum isolates associated with onion. Therefore, these characteristics were investigated in 19 F oxysporum f. sp. cepae isolates from Colorado, 27 E oxysporum f. sp. cepae and 33 F. oxysporum isolates nonpathogenic to onion from South Africa. Six E oxysporum f. sp. cepae VCGs (0421 to 0426) were identified, of which three were new. The dominant VCGs in Colorado and South Africa were VCG 0421 (47% of isolates) and VCG 0425 (74%), respectively. VCG 0423 was the only VCG that was shared between the two regions. Molecular phylogenies (intergenic spacer region of the rDNA, elongation factor la, and mitochondrial small-subunit) confirmed the polyphyletic nature of E oxysporum f. sp. cepae and showed that some F. oxysporum f. sp. cepae and nonpathogenic E oxysporum isolates were genetically related. Most E oxysporum f. sp. cepae isolates clustered into two distinct, well-supported clades. The largest clade only contained highly virulent isolates, including the two main VCGs (0421 and 0425), whereas the basal clade mostly contained moderately virulent isolates. These groupings along with the VCG data provide an important basis for selection of isolates for use in breeding programs, and for the development of molecular makers to identify VCGs. Mating type genotyping revealed the distribution of both mating type (MAT1-1 and MAT1-2) idiomorphs across phylogenetic clades, and the fact that several isolates contained both idiomorphs.

Phosphonic acid: a long-standing and versatile crop protectant

Summary: Phosphonic acid-based fungicides, or phosphonates, have been widely used as crop protectants in horticulture since the late 1970s, and recently in native ecosystems and forestry. Their effectiveness against difficult-to-manage foliar and soilborne diseases makes them crucial for protecting forests and sensitive ecosystems under threat from pathogens. Their versatility in agriculture and broader applications, along with continued research on efficacy, safety, and interactions, will support their continued use.

Summary: Citrus black spot (CBS) caused by Phyllosticta citricarpa is a global disease affecting citrus. Research in two lemon orchards in South Africa found that while sexual reproduction may be the primary mode of transmission, a significant role of asexual reproduction was also observed. Predominant genotypes were identified in both orchards causing a large percentage of fruit infections. Distance was not found to be a reliable predictor of genetic substructuring, with significant genetic differentiation observed between the two orchards.

Phosphonate Treatment Effects on Phytophthora Root Rot Control, Phosphite Residues and Phytophthora cactorum Inoculum in Young Apple Orchards

Summary: Fusarium wilt, caused by the fungus Fusarium oxysporum f. sp. cubense (Foc), is a significant threat to global banana production. The tropical race 4 (TR4) variant of Foc is highly virulent and mainly affects Cavendish bananas. Recent surveys in Laos and Vietnam show that Foc TR4 is mainly restricted to the northern regions of these countries and is limited to Cavendish cultivation settings. The disease may have been introduced through infected planting material brought by Chinese companies. Clear communication efforts on disease epidemiology and management with emphasis on biosecurity practices need to be improved to prevent further spread.

Summary: This study identified a GPI-anchored protein FocECM33 that plays a crucial role in the growth and pathogenicity of Fusarium oxysporum f. sp. cubense tropical race 4 (Foc TR4). FocECM33 was highly upregulated during the early infection process in banana roots and its disruption led to decreased hyphal growth, increased sensitivity to cell wall stresses, and reduced virulence. Transcription analysis revealed that FocECM33 influences the production of secondary metabolites and regulates biosynthetic processes in Foc TR4.

Summary: This study reveals the potential mechanism of manipulating iron bioavailability in the banana rhizosphere to suppress Fusarium wilt. The results suggest that iron starvation induces the production of iron-scavenging secondary metabolites, called siderophores, by Foc TR4 as a counteractive mechanism to evade iron starvation.
