An acyltransferase-like gene obtained by differential gene expression profiles of quinolizidine alkaloid-producing and nonproducing cultivars of Lupinus angustifolius

Quinolizidine alkaloids (QAs) are one of the representative groups of plant alkaloids. To isolate genes involved in QA biosynthesis, we performed a differential gene expression analysis by PCR-select subtraction between a QA-producing bitter cultivar and a nonproducing sweet cultivar of Lupinus angustifolius. We obtained 71 and 43 clones specific to the bitter and sweet cultivars, respectively. Among the genes specifically expressed in the bitter cultivar, an acyltransferase-like gene (LaAT: Lupinus angustifolius acyltransferase) showing homology to the BAHD protein family was isolated. LaAT showed the strongest homology to the Arabidopsis thaliana BAHD acyltransferases involved in the formation of conjugated polyamines. Semi-quantitative RT-PCR revealed that LaAT expression was highest in the young leaves but barely detectable in the other organs of the bitter cultivar plant, whereas LaAT expression was undetectable in the sweet cultivar.

Summary: The study demonstrated the fabrication of poly(glycerol adipate) nanoparticles decorated with folic acid and triphenylphosphonium and their potential for delivering acetogenin-enriched leaf extract to ovarian cancer cells. The nanoparticles exhibited small size, narrow size distribution, high negative surface charge, and a biphasic sustained release of the extract. The FOL/TPP-decorated nanoparticles showed higher drug loading, enhanced toxicity to cancer cells, and improved cellular uptake compared to FOL-decorated nanoparticles. The FOL/TPP-NPs also demonstrated better stability, uptake efficiency, and targeting ability to mitochondria.

Downregulation of a cluster of genes encoding nitrate transporter 1/peptide transporter family proteins in tomato with a mutated JRE4 transcription factor

Summary: A group of specialized metabolites called steroidal glycoalkaloids (SGAs) are produced in Solanum species and their biosynthesis is regulated by the transcription factor JASMONATA-RESPONSIVE ETHYLENE RESPONSE FACTOR 4 (JRE4). In this study, the downregulation of a cluster of genes encoding nitrate transporter 1/peptide transporter family (NPF) members was observed in a low-SGA tomato mutant with a loss-of-function mutation in JRE4. The NPF transporters, which belong to a large family of plant membrane transporters, are induced by the defense-related phytohormone jasmonate and are potentially involved in the SGA pathway.

Optimization of Magnetic and Paper-Based Molecularly Imprinted Polymers for Selective Extraction of Charantin in Momordica charantia

Summary: In this study, novel magnetic nanoparticles Fe3O4@MIPs and filter paper@MIPs were synthesized by sol-gel polymerization for selective extraction of charantin. Compared to traditional liquid-liquid extraction, these methods exhibited excellent extraction efficiency and selectivity, and were rapid, inexpensive, simple, reproducible, and environmentally friendly.

CD44-Targeted Lipid Polymer Hybrid Nanoparticles Enhance Anti-Breast Cancer Effect of Cordyceps militaris Extracts

Summary: This study aimed to enhance the anticancer effect of Cordyceps militaris herbal extract (CME) on breast cancer cells by using hyaluronic acid (HYA) surface-decorated lipid polymer hybrid nanoparticles (LPNPs), and evaluate the feasibility of using a synthesized poly(glycerol adipate) (PGA) polymer for LPNP preparation. The results showed that the synthesized polymers could successfully encapsulate CME in LPNPs. The HYA-decorated PGA-based LPNPs significantly improved the anticancer effect of CME against breast cancer cells by enhancing cellular uptake through CD44 receptor-mediated endocytosis.

Lipidome Profiling of Phosphorus Deficiency-Tolerant Rice Cultivars Reveals Remodeling of Membrane Lipids as a Mechanism of Low P Tolerance

Summary: Plants have evolved mechanisms to tolerate low P, including changing their membrane lipid composition. This study investigated the membrane lipid remodeling among different rice cultivars under P deficiency. The levels of phospholipids were lower in -P plants compared to +P plants, while the levels of non-phospholipids were higher in -P plants. Decomposition of phospholipids in roots correlated with low P tolerance. These findings suggest that rice cultivars remodel membrane lipids under P deficiency, contributing to their low P tolerance.

Their effects on nanoparticle (NP) formation, drug release, and cellular responses in cancer and normal cells were evaluated. The results showed that both PGA-TOC NPs and PGA-CHO NPs could be used as delivery systems for anticancer drugs, with different characteristics and effects on cancer and normal cells.
