Characterization of mutations in the iron-sulphur subunit of succinate dehydrogenase correlating with boscalid resistance in Alternaria alternata from California pistachio

Thirty-eight isolates of Alternaria alternata from pistachio orchards with a history of Pristine (pyraclostrobin + boscalid) applications and displaying high levels of resistance to boscalid fungicide (mean EC50 values >500 mu g/ml) were identified following mycelial growth tests. A cross-resistance study revealed that the same isolates were also resistant to carboxin, a known inhibitor of succinate dehydrogenase (Sdh). To determine the genetic basis of boscalid resistance in A. alternata the entire iron sulphur gene (AoSdhB) was isolated from a fungicide-sensitive isolate. The deduced amino-acid sequence showed high similarity with iron sulphur proteins (Ip) from other organisms. Comparison of AaSdhB full sequences from sensitive and resistant isolates revealed that a highly conserved histidine residue (codon CAC in sensitive isolates) was converted to either tyrosine (codon TAC, type I mutants) or arginine (codon CGC, type II mutants) at position 277. In other fungal species this residue is involved in carboxamide resistance. In this study, 10 and 5 mutants were of type I and type II respectively, while 23 other resistant isolates (type III mutants) had no mutation in the histidine codon. The point mutation detected in type I mutants was used to design a pair of allele-specific polymerase chain reaction (PCR) primers to facilitate rapid detection. A PCR-restriction fragment length polymorphism (RFLP) assay in which amplified gene fragments were digested with AciI was successfully employed for the diagnosis of type II mutants. The relevance of these modifications in A. alternata AaSdhB sequence in conferring boscalid resistance is discussed.

Apple Fruit Core Rot Agents in Greece and Control with Succinate Dehydrogenase Inhibitor Fungicides

Molecular identification and pathogenicity of Rhizoctonia solani and Pythium spp. associated with damping-off disease on baby leafy vegetables in Greece

Summary: In recent years, the cultivation of leafy vegetables as baby leaves has gained recognition in the global market and has attracted the interest of consumers. This study conducted a comprehensive analysis of specific Rhizoctonia solani AGs and Pythium spp. derived from baby leafy vegetables exhibiting damping-off symptoms in Greece. The results showed variations in pathogenicity among the different AGs of R. solani and identified three Pythium spp. The findings highlight the importance of understanding these pathogens in order to prevent and manage damping-off symptoms in baby leaf vegetables.

Root Transcriptional and Metabolic Dynamics Induced by the Plant Growth Promoting Rhizobacterium (PGPR) Bacillus subtilis Mbi600 on Cucumber Plants

Aggressiveness and Patulin Production in Penicillium expansum Multidrug Resistant Strains with Different Expression Levels of MFS and ABC Transporters, in the Presence or Absence of Fludioxonil

Summary: Penicillium expansum is a common postharvest pathogen of apple fruit, causing blue mold disease. Fungicide-resistant MDR strains with overexpression of efflux transporters have been selected. This study investigated the aggressiveness and patulin production of MDR strains, as well as the expression pattern of patulin biosynthesis pathway genes. Results showed that MDR strains produced more patulin but were less pathogenic, and the expression levels of patulin biosynthesis genes did not correlate with patulin concentration. The selection of MDR strains and their increased patulin production pose a serious concern for disease control and human health.

Resistance of Black Aspergilli Species from Grape Vineyards to SDHI, QoI, DMI, and Phenylpyrrole Fungicides

Summary: Fungicide applications can reduce fungal populations and influence pathogen evolution. This study examined the population structure of black aspergilli species in Greek vineyards and found widespread resistance to fungicides in A. uvarum isolates from conventional vineyards. However, A. tubingensis isolates showed sensitivity to pyraclostrobin. Sequencing analysis revealed mutations in the genes of resistant A. uvarum isolates, suggesting the contribution of fungicide resistance to population structure.
